| Paeonia lactiflora Pall,is a perennial herbaceous plant of Paeoniaceae Paeona.Paeonia lactiflora is very popular and widely used,especially in landscape landscaping and medical medicine.However,peony has a special double dormancy and germination characteristics of upper and lower hypocotyls,which leads to low germination rate,seriously affecting cultivation and application of breeding.At present,there are few studies on seed germination conditions of Paeonia lactiflora Pall,and most of them are focused on breaking down lower hypocotyl dormancy and promoting rooting.There are few literatures about how to break the dormancy of the upper hypocotyl and germinate.The germination of Paeonia lactiflora seeds in different storage years was comparatively few.Therefore,exploring the suitable germination conditions of Paeonia lactiflora seeds in different storage years has broad research prospects,and plays a key role in seed germination of Paeonia lactiflora Pall.It is of great significance for actual production.In this experiment,we thoroughly studied the biological characteristics,germination conditions and physiological indexes of Paeonia lactiflora seeds in different storage years,and explored the best conditions for seed germination of Paeonia lactiflora Pall.The results of the study are as follows:1.Biological characteristics of Paeonia lactiflora seeds.The seed germination characteristics of Paeonia lactiflora will directly affect the result of seed germination.In this experiment,the seeds of peony seed with uniform uniformity and full plump were used.The viability of non-year-storage and one-year-storage Paeonia lactiflora seeds was high,and the potential of germination was great.The viability of two-year-storage Paeonia lactiflora seeds was low and the potential of germination was weak.2.Experimental results of breaking the hypocotyl dormancy of Paeonia lactiflora seeds in different storage years.Under the same storage year,-40 treatment 30 days before rooting,geting the best rooting results and the highest rooting rate;In different storage years,under the same low temperature treatment,the rooting result of one-year storage seeds is better than non-year-storage peony seeds,which was better than that of peony seeds stored for two years.3.Experimental results of breaking the epicotyl dormancy of Paeonia lactiflora one-year-storage seeds.The comprehensive treatment results of 4 ℃ low temperature treatment with different concentrations of gibberellin were better than 4 ℃ low temperature treatment or gibberellic acid alone,and the combination treatment result of 4 ℃ for 21 days with 300mg/L of gibberellin was the best,the germination rate reached 75%and the germination index was 0.683.4.Changes of physiological indexes in rooting experiment.The water content of Paeonia lactiflora Pall.seeds in the same storage year increased continuously during the rooting process;soluble sugar content increased first and then decreased;starch content continued to decrease;soluble protein content first decreased and then increased.The activity of peroxidase(POD)increased first and then decreased,and the activity of catalase(CAT)decreased first and then continued to increase.5.Changes of physiological indexes in germination experiment.The water content during germination of one-year-storage Paeonia lactiflora seeds rose first and then;the content of soluble sugar first decreased and then increased;the starch content decreased first,then increased;the protein content decreased first and then increased in the germination process and finally reduce.For one-year-storage Paeonia lactiflora seeds,and the activity of POD increased first and then decreased during germination,and the activity of CAT showed an upward trend. |
