Rehmannia glutinosa f.Hueichingensis(Chen et Sehih)Hsiao is perennial herbs belonging to the family scrophulariaceae,one of the Chinese famous “Four Huaiqing Chinese medicines ”,widely distributed in wenxian,wuzhi,and Qinyang of Henan,etc.It has long-term vegetative propagation with tubers,serious damage to pests,resulting in decreased yield,poor quality,and degraded varieties.In this paper,Based on the previous won six "85-5" Chromosome reduplication of Rehmannia glutinosa f.Hueichingensis(Chen et Sehih)Hsiao(It is represented by At1,At2,At3,At4,At5,At6)respectively for material in the field of tube seedlings and the fifth generation plants form a comparative study on physiological,production,quality,the flood tolerance.In order to select the optimal strain in the chromosome doubling strain,The results are as follows: 1.Morphology: Plant height,number of leaves,maximum leaf area,and crown width all reached their maximum in October.There was no significant difference in the plant height of each line in the early growth stage.The chromosome doubled plants were higher in the late growth stage than in the undoubling plants,and the At6 and At5 of the strain had significant differences compared to the control.There was no significant difference in the number of leaves of each line in the early growth stage.The chromosome doubled plants were more in the late growth stage than in the undoubling plants,and the At2,At3 and At6 of the strain have significant differences compared to the control.The maximum area of the leaves and the crown were no significantly different,between doubled and chromosome doubled plants.2.Physiological and biochemical aspects: Chromosome doubled plant photosynthetic pigments(chlorophyll a,chlorophyll b,total chlorophyll content,and carotenoid content)were no significantly different from the control at all stages;the soluble protein content was higher than the control,in the late growth stage.,At3 was the highest;the total soluble sugars of some chromosome doubling plants(At5)were higher than the control and reached significant levels.3.Yield: After doubling chromosomes,there was no significant difference in yield between At2 and At5 compared with the control.The yield of other lines was lower than that of the control and the difference was significant.4.Quality: After the doubling of chromosomes,the content of verbascoside in all strains was higher than that in the control,of which At2 was the highest(0.22%),which was 7 times that of the control: the At2 and At3 furfural contents were not significantly different from the control.The sterol content of other strains was lower than that of the control and the difference was significant.5.Tolerance: From the morphological point of view,after the chromosomes were doubled,the resistance of different strains was different.At4 was more resistant,At2 was moderately resistant,and At1,At3,At5,At6 were less resistant to the control.From the point of view of stress resistance,the free proline content of the chromosome-doubling strain was higher than that of the control before 5 days,and the MDA content was lower than that of the control,indicating that the chromosome resistance was enhanced after doubling the chromosome.6.DNA methylation levels: The methylation level of the chromosome-doubling strains was lower than that of the un-doubling strains,showing the phenomenon of demethylation.In summary,At2 was selected as a high-yield,high-quality,and stress-resistant strain from six autotetraploid strains,which laid the foundation for further breeding of excellent new varieties of Rehmannia glutinosa f.Hueichingensis(Chen et Sehih)Hsiao. |
