| Pasteurella multocida(Pm)can cause pasteurellosis in a variety of animals and humans.The control of Pasteurellosis is still a difficult problem.In this study,genomes of PmA and PmD was compared to reveal the pathogenic mechanism of Pasteurella multocida.At the same time,the inhibition mechanism of santalol to PmA was studied using transcriptome method,which accum Late data for new antimicrobial development.1.Comparative genomic analysis of PmA and PmD whole genome of PmA and PmD were sequenced by Illumina Hi Seq 4000 system method.The results of comparable genomic analysis showed that PmA and PmD had the same genome size and G + C content.Genome size and G + C content of PmA and PmD was 2599048 bp and 39.932%,2274155 bp,G + C was 40.26% respectively.Genome PmA and PmD encoded 2580 genes and 2130 genes respectively.Genome of PmA and PmD shared the same metabolic pathway.The difference between the two genomes of PmA and PmD was reflected in type of some genes and the copy number of some genes.According to the analysis of evolution of Pm different serotype,the evolution of Pm was divided into two clades,that is,one clade of Pm F alone,and the other four serotypes were another clade.These four serotypes had a close phylogentic relationship.The evolution groups of different serotypes(including molecular type)had not obvious host specificity,but had some regional characteristics of geographic distribution area.2.Transcriptomic analysis on the mechanism of Pasteurella multocida resistance of alanthol The experimental results showed that santalol had a good antimicrobial effect on PmA.The transcriptome analysis showed that the expression of 2864 genes changed after Pm was treated with santalol,of which 43 genes were significantly differential expression:23 genes were significantly up regulated,20 genes down-regulated significantly.Among these genes,13 ribosomal protein genes were affected,and fts Z,pri B,rpo E and nus G were significantly down regulated.Therefore,it wae suggested thay the antibacterial effect of santalol is mainly implemented through the influence of gene duplication,translation and transcription,and also the cell division of Pm.To sum up,the genomic differences between PmA and PmD are small,including copy number and type of genes.PmA,B,D and E had a close phylogentic relationship compared to Pm F.The antimicrobial mechanisms of santalol is involved in the regulation of gene replication,translation and transcription,and cell division of Pm. |
