Tissue-specific promoters direct the expression of genes in specific tissues or organs of plants,which can avoid unnecessary waste of plant and are therefore important cis-acting elements in genetic engineering.In this study,Common Wild Rice(Oryza rufipogon Griff.)was used as the material.Nine root-specific expression candidate genes were screened by analyzing the wild rice cDNA library constructed in the early stage of the laboratory.The promoters of these genes were cloned and plant expression vector constructs containing the ?-glucuronidas(gus)reporter gene were separately constructed and transformed into Arabidopsis thaliana.The GUS histochemical staining and quantification of transgenic Arabidopsis all showed that OrRSGp regulates gus gene expression only in the root.The results of expression analysis of transformed rice were consistent with transformed Arabidopsis thaliana.Bioinformatics analysis shows that the promoter has a length of 896 bp and contains major functional elements such as CAAT-box and TATA-box,as well as cis-acting regulatory element involved in the MeJA-responsiveness and some elements involved in defense and stress responsiveness.OrRSGp is a root tissue-specific promoter that can provide new regulatory elements for plant genetic engineering.Drought is one of the most important environmental pressures to reduce crop production.The root development of plants is directly affected by environmental factors,and plants with well-developed root systems can better absorb water from deeper soils.Common Wild Rice is rich in genetic diversity and has a well-developed root system,which is characterized by its perennial breeding.Based on the cDNA library of common wild rice constructed in previous experiments,17 candidate genes were screened through bioinformatics analysis.Constructing expression vectors and transforming Arabidopsis thaliana.Through drought treatment and phenotypic analysis of transgenic Arabidopsis thaliana,it was finally determined that OrAELP 1 is a drought-resistant gene in common wild rice.The drought-resistance-related physiological indices and drought-resistance pathway genes were detected in Arabidopsis overexpressing OrAELP 1,which preliminarily proved that it enhanced drought resistance in Arabidopsis thaliana.
