A plant is a complex micro-ecological system,and its various tissues are inhabited by various endophytes.Endophytes are an important part of plant micro-ecology.In the long-term interaction with plants,they constitute a complex and diverse community structure and have important biological functions.Under the long-term natural selection,wild rice has accumulated a large number of excellent traits and is an important germplasm resource in crop breeding.To study the diversity of endophytic community structure and its influencing factors in wild rice,it is important to understand the dynamic changes and distribution of endophytes and reveal the mechanism of interaction between microorganisms and plants.This study collected wild rices from 4 continents,17 countries,6 genotypes,and 17 cultivars.The high-throughput sequencing technology system was used to systematically study the community structure and diversity of endophytic bacteria,and through biological information.A variety of statistical methods were used to study the effects of various factors on the community structure of endophytic bacteria.The results showed that wild rice contains abundant endogenous bacterial resources,which belong to 31 phylums and 78 classes of bacteria,among which are Proteobacteria,Bacteroidetes,Firmicutes,Actinobacteria and Verrucomicrobia accounted for 78.86%,10.63%,7.24%,2.72%,and 0.21%,respectively,which are the dominant bacterial populations of wild rice,but various in different samples.Statistical analysis showed that there was no significant difference in Shannon index,species evenness,and phylogenetic diversity of endophytic bacteria in wild rice of different gene types,but the species with significant differences abundance of FF genotype wild rice was found in stem tissues,it was Paenibacillus.In the root tissue,the species with significant difference in BBCC genotype wild rice was Fluviicola,and the species with significant difference in FF genotype wild rice was Novispirillum,indicating that the genotype of wild rice had no significant effect on the endogenous bacterial diversity.Among different species of wild rice,there was a significant difference between O.meridionalis and O.brachyantha,O.minuta,O.ruffipogon in Shannon index,evenness and phylogenetic diversity(p<0.05),and there were 12 species wild rice,which has the species with significant difference,especially the wild rice from Australian have the most abundant species.Shows that there are significant differences between different types of wild riceIn wild rice,the Shannon index and phylogenetic diversity of endophytes in leaves and roots,roots and stems were significantly different in different sites(p<0.001),and there were significant differences in species evenness(p<0.05).Different tissue samples in the PCoA plot clearly separated along the coordinates of the primary and secondary cylinders.And 11 species and 1 species with significant differences in abundance were found in roots and stems,all of which proved that different tissue parts are the main factors affecting the endogenous bacterial diversity.Wild rice from different countries and regions has significant differences in Shannon index and phylogenetic diversity(p<0.05).LEfSe shows that the wild rice in 9 countries from 4 continents has species with significant differences in abundance.Among them,in the genus level,wild rice from Madagascar from Africa have 37 genera with significant differences in abundance,indicating that different geographical sources maybe affect the diversity of endogenous bacteria in wild rice.Typical association analysis(CCA)showed that there was a significant positive correlation between physicochemical properties of wild rice and some bacterial communities,among which TP had the greatest impact on the microbial community structure,indicating that the physicochemical properties were also the factors affecting the endogenous bacterial diversity of wild rice.At the same time,by constructing the total abundance network of wild rice endophytic bacteria,some closely related complex flora were found,which proved that the endophyte bacteria of wild rice have complex interspecific relationships.
