Rice is an important food crop in the world.It is widely planted in the north and south of China.Drought and salinity are important factors affecting the yield and quality of rice,so the research on rice water-saving and drought resistance is very important.As a variant of rice,upland rice has extremely strong ecological adaptability.However,due to certain deficiencies in upland rice and environmental deterioration,its yield and quality are limited.Therefore,optimization of upland rice traits has become an important research direction.In the previous issue,Agrobacterium transformation was used to transfer the Late Embryogenesis Abundant Proteins(LEA)family gene DEHY into the Kunhan 1,the Handao 2 and the Hannuo 3.The transferred lines was used in progeny identification analysis and screening.In the 98 transgenic lines obtained in the previous experiments,96 homozygous and 2 heterozygous strains of the DEHY gene are picked up by PCR and herbicide resistance analysis;the result of the resistance assessment indicates that the transgenic materials resistance to drought and the salt tolerance are higher than that of the wild type.Through the herbicide limit test,96 strains of homozygous candidate strains are identified,and the highest tolerated concentration is 8 mg/mL,which is 10 times greater than the conventional experiment uses.Through field mutant screening,34 transgenic mutants,such as plump and large panicles,were successfully screened out.By Southern hybridization,one single-copy strain,two double-copy strains,and one three-copy strain were selected from the homozygous candidate strains;single-copy strains were identified by TAIL-PCR,and the results show that the exogenous gene DEHY is successfully inserted into chromosome 4 of the rice genome.Finally,single-copy homozygous lines with stable genetic traits were selected from transformed plants.
