Rice is one of the main sources of grain for people.During the period of growth,the water requirement is large for rice.The cultivation of rice is limited by the environment and water resources.Compared with the rice,the water consumption of upland rice is smaller,but problems,such as drought,weeds,saline-alkali land,have been important factors affecting the production of upland rice.In order to solve those problems,some stress resistance and herbicide resistance genes were transferred into upland rice,then phenotypic screening and molecular identification were used to obtain upland rice lines with stable inheritance and better characters.The previous trial of this project has obtained 82 PPA,APX,and 8704 transgenic upland rice.Through the screening of laboratory herbicides and PCR identification,46 transgenic candidate homozygous lines were initially screened out.The salt-tolerant and anti-drought analysis of transgenic APX and 8704 transgenic homozygous strains showed that compared with the non-transgenic strains,the salt-tolerant and anti-drought ability of the transgenic strains were improved.The herbicide limit analysis was performed on the 46 transgenic candidate homozygous strains,and the highest concentration of the herbicide resistance of the transgenic material is 8 mg/mL.By Southern blot analysis,copy number analysis were performed on transgenic candidate homozygous lines to confirm weather the single-copy line is a homozygous line.Finally,four single-copy homozygous lines were identified,which are: transgenic APX transgenic lines 6-42,transgenic PPA transgenic lines 7-32,7-40,and 7-46.Through Tail-PCR identification,the insertion site of APX transgenic single-copy line 6-42 was analyzed,and its foreign gene insertion site is on chromosome 5.
