| A new hybrid grouper(Epinephelus fuscoguttatus ♀× Epinephelus lanceolatus ♂),given its advantageous in terms of good taste,fast growth and economical value,has been regarded as a new product with potential in aquculture.So far there is not relative report about genetic structure of this hybrid grouper.To ensure its sustainable development and detect effective size of population,we decided to research the level of genetic diversity and subdivision in four geographical population of Pearl Gentian Grouper,by using microsatellite and AFLP marker.Four populations were collected from Xiangan(Xiamen,China;abbreviate to XA),Dongshan(Xiamen,China;DS),Hainan(China;HN)and Shandong(China;SD),respectively.For convenience,Pearl Gentian Grouper would be named as PGG in the following sections.First of all,we developed 19 pairs of microsatellite primer from PGG(XA),and characterised their polymorphism,showing the values of NA,PIC,Ho,HE which ranging from 2 to 6,0.062 to 0.729(mean=0.400),0 to 1(mean=0.497)and 0.066 to 0.731(mean=0.415),respectively.And then 15 primers were selected from the 19 microsatellite primers mentioned above to do further research in four populations(120 individuals totally)for calculating genetic diversity and subdivision.The percentage of polymorphic loci was 100%to XA population,93.33%to SD,86.67%to DS,and 86.67 to DS.In summary,the mean values of NA,NE,I,HO,uHE and PIC were 2.583,1.766,0.623,0.470,0.385 and 0.623,respectively.Values of genetic identity between four populations ranged from 0.8573 to 0.9459,and values of genetic distance ranged from 0.0556 to 0.1539.The ananlysis of AFLP presented that the mean value of NA,NE,I,HO,uHE were 1.461,1.416,0.319 and 0.228 respectively,and that the mean percent of polymorphic loci was 50.82%.The value of genetic identity ranged from 0.8520 to 0.9420,while the value of genetic distance ranged from 0.0600 to 0.1640.Compared the results of AFLP with microsatellite marker’s,we found that the two molecular markers,even the values of analysis data from twe markers were varied,were showing the same trend in terms of estimating the genetic diversity and subdivision level. |
