The effect of Clostridium butyricum on growth performance,blood routine,serum biochemical indexes,anti stress and antioxidant capacity and expression of key molecules signal transduction pathway in the infection of Escherichia coli were studied in this experiment.It provides a theoretical basis for Clostridium butyricum to relieve stress reaction induced by Escherichia coli and to add to the diet.This study is divided into two parts:Experiment one,A total of 20 piglets(21 + 2)old healthy of Northeast ×Berkshire were selected,they were randomly divided into 2 groups,with 10 replicates in each group and 1piglets in each replicate.The control group was fed with basic diet.The Clostridium butyricum group was fed diets containing 0.05% of Clostridium butyricum in the basal diet.The experiment lasted for 14 d,at 14 d,broken material continuous water,and blood was collected for testing.Result:Clostridium butyricum group was compared with the control group,the average daily gain and average daily feed intake increased significantly(P<0.05);the number of platelets increased with a significant difference of(P<0.05);the total protein,globulin,white ball ratio,glutamic pyruvic transaminase,glutamic acid aminotransferase,alkaline phosphatase,gamma glutamyl transferase,creatinine and urea were not significantly different compared with the control group.Experiment two,combined with the previous chapter test,at 14 d,broken material continuous water.The basal diet group and Clostridium butyricum,each is divided into two groups.And one of their groups attacked E.coli,after 24 h,blood was collected and serum was separated for detection.Slaughtered piglets then take the liver and save it for testing.Result:Stress index,lactate dehydrogenase test group III increased compared with test group I,the difference was extremely significant(P <0.01).Compared with the control group,the creatine kinase test group II had a significant decrease(P<0.05),and the test group III had a significant decrease compared with the test group I(P<0.05);serum antioxidant capacity,Compared with the control group,the MDA of the test group I decreased,and the difference was significant(P<0.05).Compared with test group I,the level of superoxide dismutase and glutathione peroxidase of test group III decreased significantly(P<0.01).Compared with the control group and test group III,catalase increased in test group I,with a significant difference(P<0.05);antioxidant capacity of the liver,the level of malondialdehyde(liver)in test group I and test group II decreased significantly compared with the control group(P<0.01).Compared with the control group,test group I had a higherlevel of superoxide dismutase(liver)and the difference was extremely significant(P<0.01).Test group III compared with test group I,superoxide dismutase(liver)decreased,the difference was significant(P<0.05);oxidative stress related gene expression,the increase of glutathione peroxidase 1 test group II was significantly higher than that in the control group(P<0.05),and the increase of the test group III was significantly higher than test group I in the test(P<0.01).Zinc copper superoxide dismutase,test group I increased compared with the control group,the difference was significant(P <0.05),test group III was significantly higher than test group I(P<0.01);the expression of key genes in the oxidative stress pathway,ERK1 test group I increased significantly compared with the control group(P<0.01),test group III was reduced compared to test group I and the difference was extremely significant(P < 0.01).NF-?B test group I increased significantly compared with the control group(P<0.01),test group III was reduced compared to test group I and the difference was significant(P < 0.05).To sum up,adding Clostridium butyricum to feed can increase average daily gain and average daily intake of weaned piglets;The stress caused by Escherichia coli infection in weaned piglets is in the serum lactate dehydrogenase,creatine kinase and oxidative stress serum antioxidant enzymes,liver antioxidant enzymes,the expression of liver(GPX1,GPX4,CuZnSOD)related genes and the expression of key molecules of oxidative stress pathway(ERK1 and NF-kappa B)played a protective role in piglets.This indicates that Clostridium butyricum has a significant effect on the growth performance of piglets,and plays a protective role in the stress response induced by E.coli.
