Eleusine Indica Primary Screening For Biological Control Microorganisms

Eleusine indica is one of common harmful weeds in the field.It is widely distributed throughout the country.Now,it is an important method to prevent Eleusine indica by using chemical herbicides.However,with the heavy using of chemical herbicides,it leads to a serious environmental pollution problem,which poses a great obstacle to the development of agriculture in the future.Hence,the research and development for microbial herbicides will be an efficient path to control weeds and replace chemical herbicides.The study mainly base on biological characteristics of Eleusine indica which will help study on screening of microbial herbicide.Study results are as follows:Roots,stems,leaves and soil samples of the disease plants of Eleusine indica were collected in Handan area along with villages.Twenty-six plants were obtained by mediumisolation and purification.Two dominant strains called T-1-3 and F-VD were acquired by sclertium separation leaf method in preliminary screening.T-1-3 and F-VD were proved to be Verticillium dahliae.These two strains have different levels of inhibiting effect on Eleusine indica.Detached leaf that was inoculated for 3 days reached 3disease stage and 5% of blade leaf diseased.Detached leaf that was inoculated for 5 days reached 5disease stage and and 20% of blade leaf diseased and leaves rotted.The results of research showed Eleusine indica could be inhibitted by T-1-3 and F-VD.Verticillium dahliae were cultured in PDA medium.Their morphologics were observed under a microscope after 7 days.T-1-3 has rugose colonies and black brown moniliform hyphaes.F-VD has rounded and loosened colonies.It can be stirred up easily.In the study,molecular biology assay is the main method combined with morphological identification.After study,Verticillium wilt fungus Dahlia?Verticillium dahliae versu fungal were identified.Verticillium wilt fungus Dahlia and Verticillium dahliae versu fungal of fermentation broth was sprinkled on Eleusine indica of 2-3 leaf stage in pot culture.The results showed that the two strians have srong pathopoiesis towards eleusine.Mobidity extent of Eleusine indica reached 6 classes.Meanwhile,three major crops in Hebei province including wheat,crop and cotton were tested in safety.The results indicated that two strains had few effects on them.They had a normal growth.
