Liliaceae plant is a major population of monocotyledonous plants,ornamental,edible and medicinal value and is widely distributed.Northeast China is the largest natural forest is the coexistence of 25 genera,88 species of wild Liliaceae,only a small part has been developed and utilized,even has been the development of the types,the number of small,the utilization proportion is very low.Climate change,natural forests make wild liliaceous plants in Northeast China growth environment continued to deteriorate.Domestication can protect wild varieties,the introduction of wild species can adapt to the growth of in vitro propagation of short time a large number of breeding offspring,more germplasm resources,increase the utilization of germplasm resources speed.It can alleviate the endangered wild resources gradually reduce the difficulties,can carry forward the unique wildlife resources in China.The investigation,collection,introduction and breeding of wild lily germplasm resources in Northeast China were carried out.The findings are as follows:1.Ten species of wild Liliaceae plants were collected in three plots in Northeast China.Polygonatum humileum,Polygonatum involucratum,Veratrum nigrum,Convallaria majalis.,Maianthemum bifolium,Lilium distichum,Paris veriticillata,Fritillaria ussuriensis.Smilacina japonica.and Lilium dauricum.In the range of 300-700 m above the sea level,most of the Lilium distichum are distributed in cool,moist place on the edge of a forest.The roots are thick and easy to be connected to the roots of the adjacent trees and shrubs.The growth level of Convallaria majalis&Maianthemum bifolium is relatively high in a cool and humid place in the environment.In the moist environment,Polygonatum humileum,Polygonatum involucratum,Veratrum nigrum,Paris veriticillata and Lilium dauricum grow under the forest with the sunny slope,Fritillaria ussuriensis,Smilacina japonica in the understory forest edge lawn furrow edge,in the wetter environment.2.Ten species of wild lily plants collected in the field have different environmental adaptability.Among them,the best survival rate of Lilium distichum to adapt to the ecological factors in introduced land was about 89.3%,and there was no significant difference between the external morphology and phenology of bud,florescence,plant height and the original land,and there was no significant difference in growth between Lilium distichum and the original land.The survival rate of Convallaria majalis with the worst adaptability was only 14.3%,and the seed germination rate of Lilium dauricum was 32%.Although the germination percentage was low,but the growth was better,it could grow well in the following spring.3.In vitro rapid propagation of two plants with good growth in introduced and cultivated species.In vitro rapid propagation of Lilium distichum,the best sterilization effect and growth condition were obtained by taking scale as explant and taking the medium scale and sterilizing agent as 75% alcohol 60 s+2% NaClO solution 12 min.The percentage of long bacteria was 15.57 and the percentage of scale necrosis was 3.95.The best combination of primary plant growth regulators was 6-BA 1.0 mg/L+KT 0.2 mg/L+NAA 0.15 mg/L+sucrose 30 g/L.The optimum culture medium combination was: basic MS medium+sucrose 50 g/L+6-BA 0.2 mg/L+NAA 1.0 mg/L,K-factor was 6.94;rooting medium combination of 1/2 MS+AC 0.25 g/L+IBA 0.1 mg/L+ sucrose 50 g/L;The root percentage was 100 and the average number of roots was 11.08;The optimum culture medium was peat: vermiculite: perlite=1:1:1.The seedling survival percentage was 85.7%.When seeds of Lilium dauricum were used as explants,the suitable disinfectant combination was 75% alcohol 30 s + 2% NaClO solution 8 min.The pollution percentage was 41.1%;the best culture medium was: MS +sucrose 50 g/L+6-BA 0.6 mg/L+NAA 1.0 mg/L,K-factor was 6.73;the best rooting medium combination was 1/2 MS+AC 0.5 g/L+IBA 0.1mg/L+sucrose 50 g/L,the rooting percentage was 100,the average number of rooting was 9.57.The best substrate for domestication and transplantation was peat: vermiculite:perlite=1:1:1.The survival percentage of tissue culture seedlings was 88.2.
