Raspberry is a kind of perennial deciduous shrub,rich in nutrition and hygienical.Raspberry industry develops rapidly in our nation.The demand of raspberry plantlet is increasing and the raspberry breeding method is attracted more attention.Raspberry root propagation is an innovative breeding method,being advantaged in easy operation,high efficiency and good quality.This technology has been reported in foreign,but it has not been applied in our nation.Raspberry tissue culture is an efficient and high quality plant breeding technology.Due to the materials,operation and environment,pollution is easy to occur in tissue culture.But there is less research on antibacterial and fungistatic technology in raspberry tissue culture.In the research,we used the"Polka","Autumn Britten"roots,"Autumn Britten"pollutional tissue culture seedlings as materials.The research applied to raspberry root propagation system and explored the best fungistatic(bacteriostatic)agent in tissue culture,optimizing the root propagation method and controlling the pollution in tissue culture.The studies aimed to solve the raspberry breeding problems and provided theoretical basis and important references for the breeding of raspberry.The results are as follows:(1)Two–years root of"Polka"and"Autumn Britten"could produce 50-60 root adventitious buds.The root buds germination process lasted 56 days,starting at 7 d and the largest increasing of the root buds at 21 d after burying.The 80%of buds were distributed in the root system within 5cm from the main stem."Polka"had a strong reproductive ability.(2)The suitable substrate burying temperature was about 18~°C and the temperature of shoots cutting was 18-25~°C.The best place of cutting the shoots was at 4 cm below the burying matrix.The best cutting matrix was turfy soil:perlite(1:1)+river sand.The two leaves-one burgeon shoot was best for cuttings rooting.(3)The cutting rooted at 7-9 d.Rooting agent IBA and IAA could promote rooting in the shoot cutting,and NAA had poor effects."Polka"was best in IBA 100 mg/L,"Autumn Britten"was best in IAA 100 mg/L."Polka"was better in rooting.(4)Raspberry root propagation was better than raspberry root cutting in reproductive efficiency,seedling time and seedling uniformity.Raspberry root propagation was 2.1 times of the root cutting on the number of root germination and the number of seedlings was 2-3 times.The root propagation need 65 d and root cutting propagation need 100-110 d.(5)Pollution would erupt after inoculating 5-10 d and the pollution rate was increased at 15-30d.Effects of the antibacterial agent would be reduced at 30 d,and the pollution rate increased again.In tissue culture,it is advisable to transfer plantlet within 30 d.(6)Among the 8 kinds of fungistatic(bacteriostatic)agent in multiplication stage,the best fungistatic(bacteriostatic)agent is S106 100 mg/L+chlorothalonil 200 mg/L.After 10 d culture,culture medium appeared mild pollution and pollution rate was 9%at 30 d.The pollution rate was13.4%and the proliferation efficient was 6.1.The carbendazim,mancozeb and streptomycin sulphate pollution control were bad.Though amixida,carben dazim-mancozeb had antibacterial effect,they had a significant inhibitory effect on the proliferation of tissue culture.
