The Millettia speciosa Champ is a plant of Leguminosae.It is dual-use medicinal plant of a medicinal and a food.At present,there are often problems such as tuber less production and lower yield during the Millettia speciosa Champ,which severely restricts the development of Millettia speciosa Champ planting industry.Therefore,in this study,the seedlings of Millettia speciosa Champ with better tuber ability were used as test materials.Transcriptomic sequencing was used to explore the mechanism of root enlargement from the perspective of molecular biology,and to establish the quality standard for Millettia speciosa Champ seedlings and increase in production will provide a teoretical basis.The main findings are as follows:(1)The tuber Millettia speciosa Champ seedlings was not obvious of taproot base expansion,the number of lateral roots was much more and most of the roots were enlarged,there was a clear swelling nodule phenomenon,there was a clear accumulation of starch;the common seedlings have taproot base expansion of 3 to 5 cm,thick 15?20 mm,lateral roots ware few and often absorb the roots,the surface was smooth,no significant expansion of nodules;paclobutrazol treated Millettia speciosa Champ can significantly increase the Millettia speciosa Champ expansion of the seedlings of the root biomass.(2)The number of lateral roots,the weight of lateral roots,and the maximum lateral roots of the tuber Millettia speciosa Champ were 3.12,4.33 and 3.03 times that of the common seedlings.There was significant difference of the number of expanded lateral roots,the fresh weight of underground parts,the fresh weight of lateral roots,the maximum diameter of lateral roots,and the dry weight of underground parts in the paclobutrazol-treated Millettia speciosa Champ seedlings and ordinary seedlings and tuber Millettia speciosa Champ seedlings.(3)It ware directly proportional to the fresh weight of underground parts of the Millettia speciosa Champ and the soluble sugar,sucrose,starch and the activity of starch synthase,starch branching enzyme and ADPG-pyrophosphorylase.The ordinary seedlings,tuber seedlings,and paclobutrazol-treated Millettia speciosa Champ seedlings had soluble sugar contents of 3.61,3.32,and 6.14,sucrose contents of 3.58,3.28,and 6.13,and starch contents of 1.52 and 1.41;The activity of starch synthase was 10.21,9.06,16.91;the activity of starch branching enzyme was 21.06,19.18,30.51;and the activity of ADPG-pyrophosphorylase was 6.16,5.54,8.85.(4)Transcriptome sequencing was performed on Millettia speciosa Champ seedlings with different tuber viability,obtained 64,036 unigenes,revealed that there were 213 genes associated with plant root growth,358 genes related to starch metabolism,and 383 genes related to hormone metabolism.Thereinto,61 genes were significantly different in expression,including the expansion protein,cellulose synthase,key enzymes for starch synthesis,cytokinins,ethylene,auxin,monocrolide,brassinolide and other related metabolic genes.(5)Select 5 genes of annotation as alpha,alpha-trehalose-phosphate sy,nthase,expansin,Cytokinin-O-glucosyltransferase,glycosyltransferase 5-like,cellulose synthase-like protein D2-like isoform XI for real-time fluorescence quantitative PCR,to verify the transcriptome data,the results showed that the expression of the 5 genes was consistent with that in the transcriptome data.
