Male sterility refers a phenomenon that the plant stamen are abnormality,it cannot produce functional pollen normally,The use of this property in production can reduce seed production costs and increase the genetic purity of seeds.The maize male sterile mutant K305ms derived from inbred lines 156 that Isolated from 60Co-y-ray induced population.Based on the previous research,K305ms is genetically stable and the sterile is controlled by a single recessive nuclear gene ms305 that is located on chromosome 2L.In this study,on the one hand,we analysis homologous relationship between ms305 and ms32 through specific primers.On the other hand,the fine mapping analysis was conducted on large F2 population of A × 156 sterile strains by SSR-BSA method and Encryption tags,then predicting ms305 candidate genes.The main results were as follows:1.There is a discovery by looking up at Maize GDB?http://www.maizegdb.org/?and ms32 literatures.ms305 is close to gene clonal ms32.Compared to gene GRMZM2G163233,ms32 has a part of deletion that including multiple exons and length in it is greater than 1.6kb.Therefore,there are 6 specific primers over all exons on GRMZM2G163233.Utilizing the markers to amplify DNA of K305.K305F and K305ms follow.Using DNA man to analysis sequence results.In short,there is no alleles among ms3 2 and ms305.2.According to the Maize inbred lines B73 sequence information that published in Maize GDB?http://www.maizegdb.org/?,and Exploiting new 463 SSR primers by SSRHunter software among SSR markers bnlg469b and bnlg1940 on maize chromosome 2,Mssr2 showed polymorphism between Fertile pool?F?and sterility pool?S?.3.The loci mapping analysis was conducted on F2 population of the offspring of K305ms sterile strains and 156 inbred line.Then screening SSR primers with polymorphism were selected from SSR markers that new development the range of bin2.07-bin2.09 on maize chromosome 2 by SSR-BSA method,acquired 12 markers from polymorphic analysis of parents firstly,screened 8 markers from polymorphic analysis of gene pools finally.Then amplifing all recessive sterile plants DNA through SSR-PCR and 3%agarose gel electrophoresis.The amplificated results are linkage analyzed by MAPMAKER/EXP 3.0 mapping software.the sterile gene ms305 was preliminarily located on long arm of chromosome 2 between umc2601 and bnlg1316,and the physical distance is 1.08M.4.There are 10 homology records through analyzing genetic structure and amino acid sequence of B73 genome by and compared it with Nr database followed that hitted 206 records in it.Consult different periods of the K305ms transcriptome data,GRMZM2G416964?NC024460.2?Expressed remarkable different expression between the group of fertility and sterility.This is a SKP1-like protein 1A according to annotation among Nr?GO and KEGG and do form a SCF compound with variable elements F-box protein.It is involved with cell cycle?signal transduction?transcription of target proteins in protein and proteasome degradation,etc.
