In order to investigate the fatty acid composition and fatty acid related gene expression profile in adipose tissues of Wanxi White geese at different ages,a total of 60 Wanxi White geese with similar body weight were selected at d 70.15 geese were randomly selected for sampling at 70,80,90 and 100 d of age,respectively.Fatty acid composition of adipose tissues were determined by gas chromatographic and fatty acid related gene(SCD1,ACSL1 and ELOVL6)expression were measured by qRT-PCR.Furthermore,in order to investigate the effect of green forage on fatty acid composition and fatty acid related gene expression,a total of 240 geese with similar body weight were selected at d 70 and randomly divided into 4 groups with 3 replicates(20 per replicate)in each group.Geese in each group fed with 0,15%,30%,and 45% green forage,respectively.Five geese were randomly selected from each replicate for sampling at 70,80,90 and 100 d of age,respectively.Fatty acid composition and fatty acid related gene expression of adipose tissues were determined.Correlation analysis were performed between fatty acid composition and related gene expression levels.1.Fatty acid composition varied in adipose tissues with ageThe proportion of unsaturated fatty acid significantly greater than saturated fatty acid,As the major fatty acid,oleic acid accounted for 45.45%~47.21%.The proportion of saturated fatty acid decreased with age,while the proportion of unsaturated fatty acid increased with age.The proportion of C16:0 and C18:0 decreased with age(P<0.0001),while the proportion of C18:2 increased with age(P<0.0001).The proportion of C16:1decreased at d 80 and then increased after 90 d of age(P=0.005),while the proportion of C18:1 increased at d 80 and then decreased at d 90(P<0.0001).The proportion of unsaturated fatty acid in subcutaneous fat exhibited significantly higher than that of abdominal fat and mesenteric fat,in versely,the proportion of saturated fatty acid significantly lower than that of abdominal fat and mesenteric fat.The proportion of C16:0and C18:0 in subcutaneous fat exhibited significantly lower than that of abdominal fat and mesenteric fat,however,the proportion of C16:1 and C18:1 was higher as compared to abdominal fat and mesenteric fat.The interactions between age and adipose tissue significantly affacted the proportion of C16:0.The highest proportion of C16:0 was in mesenteric fat at d 70.The lowest proportion of C16:0 was in subcutaneous fat at d 90.2.Fatty acid related gene mRNA expression was affected by age in adipose tissuesThe expression of ELOVL6 increased at d 80 and then decreased at d 90(P=0.046).The expression of ACSL1 and SCD1 decreased with age(P=0.001,P=0.016).The expression of ELOVL6 in liver increased with age(P=0.027).The expression of SCD1 and ACSL1 varied with age(P=0.016,P=0.001).The expression of ELOVL6 in mesenteric fat exhibited significantly higher than that of abdominal fat and subcutaneous fat(P=0.001).The expression of ACSL1 in subcutaneous fat exhibited significantly higher than that of abdominal fat and mesenteric fat(P<0.0001).The expression of SCD1 in subcutaneous fat was significantly higher as compared to mesenteric fat(P=0.008).The interactions between age and adipose tissue significantly affected the expression of ELOVL6,ACSL1 and SCD1(P=0.019,P=0.002,P=0.041).3.Fatty acid composition in adipose tissues was affected by green forage additionThe proportion of C16:1,C18:2 and C18:3 increased with feeding time,in versely,the proportion of C18:0 decreased with feeding time.The proportion of C18:1 increased at d80 and then decreased at d 90(P<0.0001).The proportion of C18:2 and C18:3 was affected by green forage addition.The proportion of C16:1 and C18:2 was affected by the interaction of feeding time and green forage addition.4.Fatty acid related gene expression in adipose tissues was affected by green forageThe expression of ELOVL6 in abdominal fat increased at d 80 and then decreased at d90.The expression of ACSL1 in abdominal fat and subcutaneous fat decreased with feeding time.The expression of SCD1 in subcutaneous fat decreased with feeding time.The expression of ELOVL6 in abdominal fat and liver decreased with 15% green forage addition and then increased with 30-45% green forage addition.The expression of ACSL1 and SCD1 in adipose tissues decreased with green forage addition.The expression of SCD1 was affected by the interaction of feeding time and green forage addition.5.Correlation analysis between fatty acid and related gene mRNA expression level.The higher ELOVL6 expression level was associated with higher C18:0 and C18:1content,while lower C18:2 content.The higher ACSL1 mRNA expression level was associated with lower C18:0 content.The higher SCD1 mRNA expression level was associated with higher C16:1 and C18:1 content,while lower C18:0 content.The proportion of unsaturated fatty acid increased with age,and the proportion of saturated fatty acid decreased with age.Fatty acid composition was different among adipose tissues.Fed 15%-30% green forage will produce the highest level of C18:2 and C18:3 in adipose tissues.Genes,SCD1 ? ELOVL6 and ACSL1,could be considered as candidate genes in regulating fatty acid anabolism.
