Inula lineariifolia Turcz.,a perennial herb from Inula L.under compositae,has been used its acrial part,named Herba Inulae in the traditional Chinese(TCM).It had good efficacy in cold cough,phlegm retention and storage,asthmatic cough and much sputum,etc.Modern pharmacological research showed that it had many kinds of biological activity,such as anti-inflammatory,anti-tumor,antifungal and so on.At present,the wild resource of I.lineariifolia.is decreasing dramatically on the verge of extinction because of the environmental destruction and human factors.With the development and utilization of Inula lineariifolia Turcz.,the shortage of wild resource will lead to short supply.Thus,we should carry out artificial cultivation technology research of Inula lineariifolia Turcz.This paper studied on germplasm resources,seed germination characteristic and priming technology,the influence of water and light on I.lineariifolia.in order to provide a theoretical basis for artificial cultivation technology.The results are as follows:(1)Based on investigation of Llineariifolia.wild resources,collected 12 typical wild populations were used as materials to measure morphological characteristic,photosynthetic parameter and chemical constituent.It aimed to provide a theoretical basis for germplasm resources evaluation.The results showed the quantity of I.lineariifolia.was rapidly decreasing,mainly distributed in Anhui province and the central and southern areas of Jiangsu province.12 populations had relatively rich morphological diversity,there were significant differences of morphological characteristic,photosynthetic parameters and chemical constituents among populations.There was positive correlation on morphological characteristics and photosynthetic rate.12 populations were divided into three-type.The three populations of Xuyi,Mingguang and Fengyang were of narrower-longer leaf,bigger biomass,better photosynthetic and higher chemical constituents.Then they were classified for a similar group.It proved that the three populations were more suitable for cultivation and promotion.(2)Study the effect of different soil moisture on the growth and quality of I.lineariifolia.The results found that 30%-35%(severe drought)and 45%?50%(moderate drought)made plant height shorter,stem diameter thin,leaf area,leaf length,leaf width and root length smaller,the flower branches and number of roots less;photosynthetic pigments,Pn and soluble protein showed a downward trend;soluble sugar,POD,CAT and MDA increased.There were less alcoholsoluble extract,total flavonoid and flavonoid compounds content in drought treatment.In the 75%?80%(moderate wet)soil water treatment,it was good to morphogenesis,photosynthetic property and chemical composition.The alcoholsoluble extract of the aerial part reached 16%,total flavonoids of the aerial part and flowers reached 13.242 mg·g-1 and 22.564 mg·g-1.When soil water content was less 50%,it would restrict the growth and chemical composition of I.lineariifolia.It was suitable to grow in sufficient soil water,In the 75%-80%soil water treatment,the growth and quality of I.lineariifolia were better.(3)Through the different light transmittance shade net to adjust natural light,explored the effect of different light intensity on the growth and quality of L lineariifolia.The results found that stem diameter was thicker,flowers and roots were more and roots were longer,photosynthetic property better,soluble sugar content and chemical component higher in natural light.The alcoholsoluble extract of the aerial part reached 19%,the total flavonoids of the aerial part and flowers reached 11.825 mg·g-1 and 36.043 mg·g-1.With the decrease of light intensity and the extension of processing time,leaf area,flowers and roots showed a downward trend,weak photosynthetic property,MDA content and CAT activity increased significantly.I.lineariifolia.was light-loving plant.Shade treatment was bad for yield and quality of I.lineariifolia.(4)Through different filter membrane to adjust light quality,explored the effect of different light quality on the growth and quality of I.lineariifolia.The results found that plant height was taller in red and yellow light;shorter in blue light;stem diameter was the thickest in red light;geringst in blue light;The chlorophyll a and b content of the leaves increased significantly in red and yellow light treatment;The ratio of chlorophyll a,b and the soluble protein content increased significantly in blue light;The soluble sugar content increased in red and yellow light;The alcoholsoluble extract of the I.lineariifolia aerial part was higher in natural light,flavonoids compounds were higher in yellow and blue light.Longer wavelengths of light was good for the growth and quality of I.lineariifolia.It had light qualitative characteristics of light-loving plant.The result was the same to light intensity treatment.(5)Analysis on the seed characteristics and germination characteristics of I.lineariifolia.,studied the effect of different initiator on the seed germination.The results showed that the seed was very small with white pappus,1000 grain weight was about 0.08 g,the seed vigor was weak,the embryo developped partially.25 ? temperature were more suitable for the germination of the seed,the germination rate reached from 15%?20%;The best soil matrix was sandy soil;the germination rate reached about 16%.The initiator could promote seed germination and seed vigor.The seed priming in 5%PEG resulted in significant increase in germination rate and germination index.200 mg-L"1 GA3 and 0.5%CaCl2 also promoted germination rate,but it was inferior than 5%PEG.
