Owing to abundant tea polyphenol(TP),tea has the efficiency: Weight control and lipid lowering,improving immunity.In recent year,tea powder is used as the feed additives,has attracted widespread social attention to its efficacy.This researching experiment is to choose 8“Duchangda”(i.e.Duluoke pig,Changbai pig,and Dabai pig)three-way crossbreeding hogs,to divide into two teams: control group and test group at random,some swine are fed with basal ration in control group,others swine are fed with basal ration added 2% tea powder in test group.After the experimental stage of 3 month,studying the back-fat thickness of hog,serum biochemical indices and meat quality influenced by the tea powder;By means of RNA-Seq technology,analysizing the difference of the related gene expression of the above-mentioned two groups and linkage of Pathway enrichment,and exploring the genetic system that tea powder impacts on hog’s fat deposition.This result shows:(1)In the latter group,adding tea powder,the hog has backfat thickness,significant decrease muscle fat content(P<0.05),and has no obvious change about muscle protein content,which show that additive of tea powder may lower the fat sedimentation of hog’s carcass;(2)The blood total cholesterol and triglyceride in the latter group obviously much lowers than those(P<0.05)in the former group,however,the Cholesterol of high-density lipoprotein and low-density lipoprotein has no obvious change(P>0.05),and blood protein components also has no significant change(P>0.05).(3)Adding some tea powder in the daily ration,the genetic expression level of THRSP is clearly drived down.The analysis of RNA-Seq show shat the two groups exists 1328 gene differential expression,620 gene down-regulation,708up-regulation;The genetic expression change of THRSP、ApoB and ABCA1 are closely related to hog’s fat deposition.Analysizing the GO enrichment and KEGG pathwaw,you can find that fatty metabolism and fat digestion play an important role in the process of fat deposition after absorbing 2 pathwaw.(4)In fat digestion and absorption pathway,tea powder group of pig scavenger receptor 36(CD36),phosphatidic acid phosphatase(PAP),microsomal triglyceride transfer protein MTP,apolipoprotein B-48(ApoB-48)ATP,ATP binding cassette subfamily A member 1(ABCA1)and apolipoprotein A-I(ApoA-I)etc.the gene expression level was significantly decreased.In lipid metabolism pathway,tea powder group of pig long chain acyl CoA dehydrogenase(ACADL),the expression level of alcohol dehydrogenase,1-alkane monooxygenase genes were down regulated and long chain acyl CoA synthetase(ACSL),long chain fatty acid CoA ligase(ACSBG),O-carnitine palmitoyl transferase(CPTIA),1 acetyl CoA oxidase,long chain acyl CoA dehydrogenase,CoA dehydrogenase,etwo acetyl acyl CoA dehydrogenase,acetyl CoA acyltransferase 1 gene expression level was increased. |
