The research objective is Clausena indica(Datz.)Oliv in Congzuo city,Guangxi.The aim of this research is to understand present distribution of Clausena indica(Datz.)Oliv in Congzuo city,and to speed up breeding and selecting the new varieties.This research has collected,identified and selected germplasm resources of Clausena indica(Datz.)Oliv.Using ISSR molecular markers and GC-MS techniques,genetic diversity and volatile oil ingredient has been analyzed.Top-grafting test and efficient cultivation technology has been studied..The main results are as follows:1.Through study the current resources of Clausena indica(Datz.)Oliv,the results show that in Chongzuo city most of the resources are wild or semi-wild,with limited varieties.Longzhou county has the most resources of Clausena indica(Datz.)Oliv with approximate 77,000 trees.A germplasm nursery has been established by collecting 50 seeds of Clausena indica(Datz.)Oliv from different areas.Four pieces of fine individuals has been selected to conduct comparative analyst with plant and fruit morphology.2.50 collected samples has been analyze their genetic diversity by using ISSR molecular markers techniques.The results show that the average percentage of polymorphic is low,only 55.15%;This study conducted clustering analysis of 50 samples divided into two groups,and within each group two sub-groups has formed.The PCA(principal component analysis)proves the dependency of classification foundation and geographical position.It has lowest polymorphism comparing with other Rutaceae genus.Gene exchange of Clausena indica(Datz.)Oliv is blocked because of isolated location,which leads to less diversity of DNA inheritance.3.Taking Guiyan 15 as test materials,grafting time and grafting method was studied and crown left allowance and scion ripe degree affects survival rate.Results show that it is better to leave a quarter of crown and half-ripe twigs by adopting cleft grafting to make a scion combination in 1-3 months.4.GC-MS detection on volatile oil of flesh,peel and leaf of Guiyan 15 shows the huge chemical differences between different areas.Compound type takes monoterpenes and its derivatives as the principal thing.It is found that there is obvious difference at volatile oil composition and its content among flesh,peel and leaf.5.The paper studies the cultivation techniques of Clausena indica(Datz.)Oliv with the training and pruning,and fertilizer as a starting point,which indicates training and pruning has the highest flower formation rate with the open centre mode,optimal comprehensive quality.As a result,training and pruning technology,scientific fertilizer are the key content of high yield and quality.
