Anthurium andraeanum is a perennial herb of the genus Araceae.It is native to the tropical and subtropical regions.Therefore,it is not resistant to low temperatures.It like moist environment and is not resistant to drought.Anthurium,with its unique phenotype and colorful flames,has been popular all over the world since it was introduced from southwestern Colombia in 1896.Nowadays,as an indoor ornamental flower,anthurium is more and more present in various families,so research on improving its drought resistance is becoming more and more important.In this study,Piriformospora indica as a Beneficial fungus to improve plant stress resistance.ABA reaction as a central pathway for stress signal transduction in plants and stress resistance.According to the results of molecular markers of multiple Anthurium varieties in the early stage.Two cultivars with relatively distant relatives in the same group were selected,and the cloning and quantitative expression analysis of MYB transcription factors in the drought-stressed ABA-dependent pathways were established by establishing the anthurium-P Symbiosis system.Therefore,the molecular mechanism of P.indica to enhance the resistance of anthurium was provided,which provided an important theoretical basis for the breeding practice of anthurium and anti-stress.The main findings are as follows:Genetic diversity and genetic relationship of 56 species of Anthurium andraeanum were analyzed by ISSR molecular marker technology: Ten polymorphic primers were screened from 100 primers and 99 bands were amplified,of which 95 were polymorphic and the polymorphic rate was95.96%.The results of the analysis using POPGENE were: nei,s gene diversity index H is 0.3036;Shannons information index I is 0.4585.All these indicate that the genetic diversity of Anthurium is high.According to the cluster analysis chart,56 Anthurium varieties were divided into 12 categories,and the results of cluster analysis showed that the ISSR analysis results had a large correlation with the color of the sepals.And the ISSR molecular marker technique can be applied to the identification of Anthurium varieties.Princess Alexia Violet,Fiorino,Sumi,Caixia,Thunderbird Luxury,Black Queen and King have unique traits,They have a long genetic distance from each other and can be used as a female parent for new varieties breeding and variety improvement,and can also be preserved as an excellent germplasm resource.2.A P.symbiotic system was constructed.The piri slowed down the leaf wilting speed to some extent.With the deepening of drought stress,the ABA content in the leaves of Anthurium andraeanum increased continuously,and the ABA accumulation in the leaves began to increase rapidly during the moderate stress."The accumulation of ABA in "Viduo"is always higher than that in“ Pink Chimpion”,which,to a certain extent,suggest that the "Pink Champion" have a higher ability of droght risistance than "Vito".Compared with the control group,the ABA content in the leaves of P.sinensis was higher than that in the control group.3.The primers were designed using primer 5 with reference to the drought stress related MYB transcription factor gene sequence already reported on NCBI.By cloning of homologous genes,two drought-stressed ABA-dependent pathway-related MYB transcription factor gene fragments were named as Aap MYB1 and Aap MYB2,which were used for subsequent quantitative expression analysis.Bioinformatics analysis showed that Aap MYB1 and Aap MYB2 genes contain a typical MYB transcription factor domain,Aap MYB1 and Aap MYB2 protein sequences have high homology,which suggest that Aap MYB1 and Aap MYB2 proteins have similar protein structure,presumably they may performe a similar function in the anthurium.4.The expression of Aap MYB1 and Aap MYB2 was determined by q RT-PCR.The expression of Aap MYB1 was not significantly different,and the expression of Aap MYB2 was significantly different-Down-regulated expression,it is speculated that Aap MYB1 gene may be involved in the drought stress response pathway independent of ABA regulatory,Aap MYB2 participates in ABA-dependent response pathway,and negatively regulates ABA.
