Traumatic is a pathological injury of animal body surface which shows trauma,burns,boils,sores,chicken pox,ulcers and so on.Wound healing has a positive meaning to animal physiology recovery.Wound healing is a complex process which involved in the inflammatory response of cells,tissue repair and the complex process of cytokine secretion and regulation,many traditional medicine and its components can affect this process,such as many kinds of traditional medicine which are having a myogenic,stasis,anti-bacterial and anti-inflammatory can promote the procedure of wound healing.At the same time,the different forms is very important to the quality of animal wound healing,especially for the topical dosage form which is very suitable for the surface wounds.These formulations are mainly creams,powders,gels,Babu agent,agent and coating agent,etc.,but the coating agent is suitable for the wound healing treatment of animals particularly.Film is made by screening the main drug,transdermal penetration enhancers,film material and the substrate.using the film,it can be short-term self-filming,and it will continue to release the drug to the wound,and the film do not need to have long-lasting and external dressings.But a good film must resolve the key issues which are the dissolving of main drug,screening of transdermal penetration enhancers,matrix composition and stable and effective,and other.Curcumin is an traditional medicine extract which have effect of antibacterial and anti-inflammatory,and wound inflammation,but the forms are mostly ointments,tablets,powders,microemulsions,have not the form of film.This study will screen the permeation enhancers which can effectively promote curcumin transdermal,determine the main drug,the most suitable proportion of transdermal penetration enhancers and the substrate,and preparation methods,carry out on skin irritation and stability test,evaluate the effect of bacterial anti-inflammatory agents and wound healing,measure the absorption and distribution of curcumin which is using after transdermal administration.These are provided the basis for preparating the film which is effective and reliable,secure and stable.Experiment ? Menthol PEG-400 in vitro percutaneous curcumin Efficacy By comparing different concentrations of menthol and different concentrations of PEG-400 on the growth of wound healing in vitro transdermal drug effects in mouse skin,screening the best penetration enhancers and their concentrations.Transdermal intelligent tester,select the mouse skin,different concentrations of menthol and PEG-400 alone and when combined with the use of percutaneous absorption of curcumin dose-effect relationship,calculated by the cumulative amount within 12 h and permeability coefficient.The results showed that when used alone,PEG-400 to promote higher cumulative amount of curcumin and its transdermal permeability coefficient,of which 6%of the PEG-400 and promote the highest cumulative amount of curcumin transdermal permeability,significantly higher than the other groups,indicating transdermal best;and menthol,and PEG-400 when used in combination,4%of menthol and 6%PEG-400 work together to promote the transdermal absorption curcumin,the highest cumulative amount of its penetration through the system,a very significantly higher than the other groups,thus determine the best plan Transdermal curcumin.Experiment ? The study of coating technology of reparation of curcumin film On the basis of utilizeing menthol as promoting transparent agent in CUR film,choosing film-forming material by orthogonal test,and the optimization of CUR film composition.With polyvinylpyrrolidone K-30(PVP K-30),ethyl cellulose(EC)and polyethylene glycol-400(PEG-400)as the film-forming material,film coating,design orthogonal experiment of comprehensive evaluation of the film coating film and film appearance.The results showed that the optimal proportion of film coating film forming materials:PVP K-30 2g,EC 1g,PEG-400 6ml,accordingly,determine the best combination of the temperture by this.At the same time,the preparation of CUR film and the film coating quality inspection are done in the excitant and stability index.The results showed that the CUR made of film coating for skin,stability,conform to the requirements of the quality of film coating.Experiment ? Efficacy of curcumin film promoting animals surgical wound healing Objective To explore the antibacterial and inflammatory of curcumin film and its effect of promoting wound healing in animals.Method In animal wound common Staphylococcus aureus,Pseudomonas aeruginosa bacteria are four indicator bacteria,bacteriostatic test was done by plate method to evaluate different concentrations of curcumin solution inhibiting wound four common bacterial infection antimicrobial effect.The acute inflammatory anti-inflammatory effect of curcumin film in mouse ear induced by xylene was evaluated by swelling test.Establish linear and circular wound model in the skin of New Zealand white rabbits,and medication on the wound model,Respectively,invasive non-drug group,Amikacin group and film treatment group were compared,at 1,3,5,7,10 and 14d after injury the wound were visually observed and assessment the effect of wound healing.the control group,the treatment group and the control group were compared,at 1,3,5,7,10 and 14d after injury the wound were visually observed and took the first 7d,14d skin around the wound tissue sections for pathological examination.Results Curcumin against Staphylococcus aureus,Pseudomonas aeruginosa and other bacteria have antibacterial effect on four,and the inhibitory effect on Staphylococcus aureus in dog is best.Ear swelling tests in mice showed that curcumin film has a certain anti-inflammatory effects.the test of the effect of curcumin film on artificial wound healing balm in rabbit results showed that wound healing of film treatment group was significantly better than the others,Pathological examination showed that the skin of film treatment group has be intact healed,it has stratum corneum on the skin,collagen fiber bundle more closely,Capillaries was more than in the control group and number of fibroblast substantially was less than the control group.Epithelial of Amikacin group has defect,the Collagen fibers of arrangement is not natural and the number of fibroblasts was substantially more than the film treatment Group;however,the invasive non-drug group secretes large amounts of fibroblast,collagen fiber bundles was loose,large gaps and uneven arranged.Experiment IV Pharmacokinetic determination of CUR film in animals Studies of CUR film in the pharmacokinetic of CUR in rats.After giving experimental rats to film coating,tested 24 hours of curcumin in rat blood drug concentration by the high performance liquid chromatography(HPLC),and analyzed pharmacokinetic parameter by non-compartment mode winnonlin 6.2.0.The results show that the rat blood drug concentration peak Cmax is 555ng/ml,time to peak Tmax isl2.4h,drug elimination half-life in plasma t1/2 is 40h,while blood drug concentration peak Cmax is 528.00ng/ml,time to peak changed to 4h and drug elimination half-life in plasma t1/2 also changed to 19.63.The test results show that the film of CUR can through the skin into the bloodstream,and maintain plasma concentration.
