Rice is one of China's major food crops,hybrid rice breeding to improve the yield of rice made a great contribution.,hybrid rice breeding depends on the application of male sterile material.The laboratory has been isolated a recessive nuclear mutant ms92 from the indica rice,it's tissue sections analysis showed that the mutant anther tapetum abnormal degradation,microspores dysplasia,and ultimately lead to infertility.This research use yeast two hybrid technology screening the interacting proteins with MS92 from rice growth period CDNA library.the corresponding T-DNA insertion mutant phenotype analysis showed that candidate genes were isolated and verified,and the results obtained are as follows:1.we screened 15 interacting protein with MS92 by yeast two-hybrid,bioinformatics and expression profiling and full-length gene validation in yeast body results show that,OsEMF2b(LOC_Os09g13630)and LOC_Os09g33670 gene are the targeted candidate genes.2.OsEMF2b(LOC_Os09g1630)gene T?DNA insertion mutants osemf2b co-separated,and the results show osemf2b mutant phenotype eo-segregated with T-DNA insertion,homozygous mutant plant is short,and the spikelets showed fewer anthers,less hull,half shell,lemma malformations such as different types of abnormal flower development;tissue sections showed that the tapetum of Mutant developmental abnormalities in microspore developmental and cavitation stage period,eventually results in the decrease of pollen sterility,even infertility.3.LOC_Os09g33670 gene T-DNA insertion mutants not only mutant phenotype co-segregated with T-DNA insertion,And hybrid mutant pollen sterility significantly reduced,and because of T-DNA insertion position is located in the promoter region of the gene-2bp place,resulting in down-regulation of gene expression.
