In this study,the deep eutectic solvent as extraction solvent were used to extract the main anthraquinones compound(aloe-emodin,rhein,emodin,chrysophanol and physcion)in Rheum palmatum L.,By the extraction rate of five anthraquinones compounds as indicator,the optimum deep eutectic solvents are screened from the 13 kinds of deep eutectic solvents and chloroform by means of principal component points analysis and the effects of the deep eutectic solvent of group points ratio,water content on the anthraquinone compounds extraction rate.at the same time,the optimized extraction conditions by the response surface optimization method.In the end,the process for purification of the anthraquinones compounds in Rheum palmatum L.by macroporous adsorption resin.The main research results are as follows:1.A HPLC-UV method for determination of aloe-emodin,rhein,emodin,chrysophanol and physcionwas developed as follow:The chromatographic column:Agilent Eclipse XDB-C18(4.6×250 mm,5μm);flow rate:1.0 mL/min,column temperature:30℃ and detection wavelength:264 nm;njection volume:10 μL;mobile phase:0.10%Phosphoric acid water(A)-Methanol(B).The following gradient duffonprogram Was used for separation:0~15 min,70~100%B;15min~20 min,100~70%B;20~25 min,70%B.2.By the extraction rate of five anthraquinones compounds as indicator,the optimum deep eutectic solvents are screened from the 13 kinds of deep eutectic solvents and chloroform by means of principal component points analysis and the effects of the deep eutectic solvent of group points ratio,water content on the anthraquinone compounds extraction rate.The optimum Deep eutectic solvents was as follows:Deep eutectic solvents(DESs): System Lactic acid:Glucose Lactic acid/Glucose acid ratio 1:2 DESs water ratio 20%3.A range of based DESs with different solvents to Lactic acid mixing ratioswere used for extraction.Other factors,such as temperature,time,water addition and liquid/soid ratio,were examined systematically using a responsesurface methodology(RSM).To select the optimal method for extracting flavonoids in Rheum palmatum L.and optimize the extraction parameters.The optimum extraction process was as follows:Microwave-assisted extraction method:extraction temperature 82℃ liquid/soid ratio 26.0 mL/g extraction time 1.5h4.The optimum process for the purification of anthraquinones compound(aloe-emodin,rhein,emodin,chrysophanol and physcion)in Rheum palmatum L.from DESs was obtained firstly.Anthraquinones compound(aloe-emodin,rhein,emodin,chrysophanol and physcion)in Rheum palmatum L.by macroporous adsorption resins was studied and the optimum adsorption and desorption parameters on the optimal DM130 resin were obtained.
