Objective:Stress is a kind of adaptive response of organism caused by the external stimulus,especially the occurrence of oxidative stress has often given to livestock and caused serious economic losses.Hericium erinaceus polysaccharide,which is extracted from the Hericium erinaceus,providing with hepatoprotective antitumor,antioxidant,hypoglycemic,hypolipidemic,anti-inflammatory,and improve immunity function,can relieve injury of oxidative stress on weaned piglets.Lipopolysaccharide(LPS)is the pathogenic composition of cell wall in the outer membrane of gram-negative bacteria,which is used to cause oxidative stress injury.In this study,LPS was used to establish oxidative stress models for weaned piglets and IPEC-J2 cells to explore the protective effect of HEP on weaned piglets induced by oxidative stress.Methods:45 healthy Duroc×Landrace×large white weanling pigs(average age was 28+3d and average weight was about 10kg)were randomly divided into 5 groups to be used to generate a control group(with basic diet),a model group(with basic diet)and three HEP groups(with 0.2%,0.4%and 0.6%HEP respectively).At the 21th d,every piglet in model group and HEP groups were injected of 100?g·kg-1 BW LPS,meanwhile,every piglets in control group were injected with equal dose of saline(NS).Blood samples were collected at 0 hours,1.5 hours,and 3 hours,then the piglets was all slaughtered to collect intestinal tissue samples,Items to be test included growth performance,serum antioxidant indexs(T-AOC,SOD,CAT,GSH-Px),serum biochemical indexes(Glucose,triglyceride,aspartate aminotransferase,alanine aminotransferase),serum immune indexes(IgA,IgG,,IgM),intestinal permeability indexes(DAO,GLN),intestinal morphology,the mRNA and protein expression of intestinal tight junction genes(ZO-1,Claudin,Occludin).Finally,the oxidative stress model of porcine small intestinal epithelial(IPEC-J2)cells was constructed by LPS to To evaluate the effect of HEP on the proliferation of IPEC-J2 cells and the expression of TJ related gene.Results:(1)0.4%HEP can promote the weaned piglets growth performance in the early weeks;(2)Basal diet added with 0.2%?0.4%?0.6%HEP can significantly increase piglets serum T-AOC,SOD,CAT and GSH-Px activities(P<0.05),significantly decrease MDA content and LDH activity(P<0.05);significantly decrease glucose and triglyceride contents,and aspartate transaminase,alanine aminotransferase and alkaline phosphatase activities(P<0.05),significantly increase serum IgA,IgG,IgM(P<0.05),significantly decrease C3,C4 contents(P<0.05),significantly increase glutamine activity(P<0.05),significantly decrease diamine oxidase(P<0.05)under the oxidantive stress induced by LPS;(3)Basal diet added with 0.2%?0.4%?0.6%HEP can relieve the morphology injury of piglets intestinal,and significantly improve the duodenum,jejunum,ileum villus height and V/C value(P<0.05)caused by the stress of LPS;(4)Basal diet added with 0.2%?0.4%?0.6%HEP can significantly inhibit the reduction of intestinal epithelial cells TJ(ZO-1,Claudin,Occludin)mRNA and protein expression caused by the stress of LPS(P<0.05);(5)HEP with different-concentrations and treating time can improve normal IPEC-J2 cell proliferation significantly(P<0.05)and alleviate the oxidative stress induced by LPS(P<0.05).The optimal concentration and treating time for HEP are 250 ?g/mL and 4 hours.In addition,HEP(with 250 ?g/mL and 4h or 8h)can significantly increase the Occludin?Claudin and ZO-1 gene expression in oxidative-stressed IPEC-J2 cells(P<0.05).Conclusion:HEP can promote the growth performance of weaned piglets;can alleviate oxidative damage caused by LPS,enhance the antioxidant capacities and immunities of weaned piglets;reduce the injury of intestinal barrier function under the stress of LPS,protect the integrity of intestinal morphology in weaned piglets,promote the expression of intestinal tight junction related genes under the LPS stress,furthermore HEP can promote the expression of IPEC-J2 cells tight junction related genes under LPS oxidative stress,to alleviate the damage of IPEC-2 barrier function induced LPS oxidative stress.
