Anoectochilus roxburghii,belongs to Orchidaceae Anoectochilus plant,it has the reputation of "Medicine King","Golden Grass" and so on.Particularly rich in polysaccharides,trace elements,organic acids,cardiac glycosides,amino acids,flavonoids and other ingredients,Anoectochilus formosanus widely planted in Taiwan,Fujian and other provinces and Southeast Asia for its,can be used in the treatment of hypertension,diabetes,heart disease,nephritis and tumor embolism.In this study,based on the studies of Anoectochilus hormone(IB A,NAA)residues,the optimum process of polysaccharides,total flavonoids and other functional ingredients,we analyzed the variation of contents of hormones and polysaccharides,total flavonoids and other functional ingredients during the growth period(0-6 months)of the artificial cultivated Anoectochilus after transplanted into the soil,this provided reference for Anoectochilus planting and rational development and utilization.1 Used High performance liquid chromatography(HPLC)to detect the hormone(IBA,NAA)content of Anoectochilus and measured the change of the hormone residues in cultivated Anoectochilus during growth.The best chromatography condition of indole butyric acid determination in Anoectochilus is V(methanol):V(water)= 80:20,mobile phase flow rate of 1.0 mL/min,column temperature 30?,detection wavelength 221 nm;Optimal chromatography condition in naphthylacetate Anoectochilus measured:V(methanol):V(water)= 60:40,mobile phase flow rate of 0.9 mL/min,column temperature 25 ?,detection wavelength 276 nm.Then separated and determined the indole butyric acid residues and naphthylacetate in Anoectochilus from 0 to 6 months.The results showed that the residues of hormones gradually reduced with the growth of Anoectochilus,the content of IBA in Anoectochilus has been reduced to 0.0019%and the content of NAA in Anoectochilus has been reduced to 0.0003%,after planted 6 months.2 Study on the extraction process and dynamic change of polysaccharides functional components of the cultivated Anoectochilus in the growth period(0-6 months).Compared with the 3 methods about water solvent extraction,microwave-assisted extraction of ethanol and ultrasonic-assisted extraction that under optimizing their optimum conditions and the same sample with the repeated trials,method of microwave-assisted extraction of ethanol that content of polysaccharide is the highest in Anoectochilus,and also is the most suitable method for the extraction of polysaccharides functional components,which has the advantage of high extraction rate and short extraction time.The optimum condition to the method of water solvent extraction to extract polysaccharide in Anoectochilus was:2 hours soaking time,water bath temperature of 60 ?,solid-liquid ratio of 1:10.The optimum condition to ultrasonic assisted extraction:50 minutes' ultrasonic time,ultrasonic temperature of 80 ?,ultrasonic power of 210 W,solid-liquid ratio of one seventy.The optimum condition to microwave-assisted extraction:3 minutes' microwave time,microwave power of 450 W,solid-liquid ratio of 1:30.The dynamic variation rule of polysaccharides of cultivated Anoectochilus in growth period:it was gradually increased first and then decreased gradually,and reached the highest after 3 months' growing.3 Study on the extraction process and dynamic change of total flavonoids functional components of the cultivated Anoectochilus in the growth period(0-6 months).Compared with the 3 methods on soxhlet extraction,ultrasonic assisted extraction and microwave-assisted extraction under optimizing their optimum conditions and the same sample with the repeated trials,method of microwave-assisted extraction that content of total flavonoids in Anoectochilus is the highest,and also is the best suitable method for the extraction of total flavonoids in Anoectochilus.The each methods of optimum conditions are as follows:the optimum conditions of the methods to the Soxhlet extraction to extract total flavonoids in tissue-cultured Anoectochilus are:ethanol volume fraction of 80%,solid-liquid ratio of 1:80,extraction time of 2.5 h;the optimum conditions to the methods of the ultrasonic assisted extraction to extract total flavonoids in tissue-cultured Anoectochilus are:ethanol volume fraction of 70%,solid-liquid ratio of 1:20,ultrasonic time of 40 min,ultrasonic power of 180 W;the optimum conditions to the methods of microwave-assisted extraction to extract total flavonoids in tissue-cultured Anoectochilus are:microwave power 300 W,the volume fraction of 60%ethanol,solid-liquid ratio of 1:30,extraction time of 5 min.The dynamic variation rule of total flavonoids of cultivated Anoectochilus in growth period(0-6 months):it was gradually increased first and then decreased gradually,and reached the highest after 4 months,growing.4 In conclusion,the proposed selection can be drawn that cultivated Anoectochilus as finished products for sale after growing 4 months is not only avail for ensure the edible safety of consumer,but function itself largely while reduce farmers planting cost.
