Keep-alive-without-water of fish transport is through gradient cooling method to bring fish into dormant state,fish body is under sealed package of CA without water after it is drawn from water environment,transfer the body into proper low temperature water environment and give a gradient warming to bring it slowly into living state after low-temperature transport.This method can greatly increase carrying capacity,improve survival rate and safety,markedly reduce cost and so on.In this research loach and crucian are made as test material,and the time and survival situation in their keep-alive-without-water is observed under different temperatures.Variations of physiological and biochemical indicators of loach are researched in the process of keep-alive-without-water and when it is subject to low-temperature stress,and the influence of cold acclimation to crucian is explored by the research method of proteomics.The research result of keep-alive-without-water and low temperature stress of loach indicate:to keep loach alive without water under three temperatures of 0?,5? and 10?,the survival rate of each group is 70%?100%and 40%at 7th day.Among them the survival rate is highest under 5?;Each group of muscle glycogen decreases gradually in the process of keep-alive-without-water,and the content of muscle glycogen is markedly lower in 5? group than in 0? group;The content of serum cortisol in each group takes on the trend of firstly-increasing-then-decreasing,with 5?group and 0? group very close to each other.All the content of glutamic-pyruvic transaminase GPT,glutamic-oxalacetic transaminease AST and lactic dehydrogenase LDH in each group has dramatic changes.Glycogen drops rapidly after loach is subject to low-temperature stress,in 8h it drops to the lowest 3.21 mg/g,then rises slowly to a stable status;The content of cortisol rises rapidly to the highest 254.31 mg/g,drops to the lowest 58.16 mg/g at 4th hour,then rises slowly until 16h and begin to drop gradually to a stable status.The physiological and biochemical indexes of serum contrast dramatically(P<0.05)compared with the control group.The research results of keep-alive-without-water and cold acclimation of crucian indicate:to keep crucian alive without water under five temperatures of-1??2??5??8? and 11?.Survival rate remains 100%when keeping alive for 54h under 2?,which is the best keep-alive-without-water temperature;When keeping alive without water after cold acclimation at 1th day,the survival rate reaches 100%high when keeping alive for 84h;The content of muscle glycogen does not contrast dramatically in each acclimation group(P>0.05,the content of cortisol contrasts dramatically(P<0.05),Its content drops to the lowest(146.8±10.95)mg/g when cold acclimation is in 1d;Whereas the content of alkaline phosphatase(ALP)and cholinesterase(CHE)does not contrast dramatically(P>0.05);The content of glutamic-pyruvic transaminase(GPT)does not contrast dramatically when cold acclimation is at 0th day and 1th day,but contrasts dramatically at 3th day and 5th day(P<0.05);All the content of glutamic-oxalacetic transaminease(AST)in each group is higher than that before the test,and contrasts dramatically when acclimation is at 0th day and at 3th day;All the content of lactic dehydrogenase in each cold acclimation group is higher than that before the test.The supporting equipment for keep-alive-without-water of fish involves temporary breeding in every aspect of transport,which effectively solves the existing problems in present equipment.The designed equipment mainly include:temporary breeding drain-pipe,fish testing fixator,transport cushion of keep-alive-without-water of fish and transport equipment of low temperature keep-alive-without-water of fish.
