As a new color of vegetables,purple cabbage(Brassica campestris L.ssp.chinensis(L.)Makino var.communis Tsen et Lee)was rich in nutrients,and could make a salad,therefore,it was deep the favor of people.Meanwhile,its anthocyanin could be used as a kind of superior natural edible pigment,thus it has great potential for application in food,cosmetic and medicine.Furthermore,light,temperature,water and other ecological factors effected the plants for normal growth and development.Study how to make scientific and reasonable regulation of purple cabbage growing conditions,was of great significance to improve the quality and production.For that two reasons,this research taked "ziwei" purple cabbage leaf as materials,firstly,to optimize the ultrasonic extraction method for anthocyanins,afterwards,and then to study the light quality,drought treatment,chilling treatment how to effect the physiology and photosynthesis of purple cabbage by manual control different ecological condition.The results were showed below:1.The ultrasonic extraction method for anthocyanins was got from purple cabbage leaves.The single factor experiment affecting the extraction yield of anthocyanin were studied,including the ethanol concentration,pH value,iltrasound power,solid-liquid ratio,time,temperature and extracting times seven factors.Then Optimized the combination experiment by central composite(Box-Behnken)and response surface methodology(RSM).In conclusion,the optimum process for anthocyanin extraction was 1:16 of solid to liquid with pH 2.76 extracted at 58? for 12 min under 70 W ultrasonic power.Under this condition,the extract yield of anthocyanins from purple cabbage leaves reached up to 4.11 mg g-1 DW.2.The quality and photosynthes of purple cabbage was affected significantly by different light quality treatment.(1)Under the B treatment,the content of anthocyanin,flavonoid,soluble protein and vitamin C were higher than the other treatment.The soluble sugar contents reached the greatest under R treatment.The light quality of RBY treatment promoted anthocyanin,flavonoid,soluble protein,vitamin C and soluble sugar content;(2)Under the RBY treatment,the value of Ci,ABS/RC,TRO/RC,ETO/RC,DIO/RC,TRO/CS,DIO/CS and MO were lowest.And that the photosynthetic pigments content and the value of Pn,Tr,Gs,PIABS,ETO/CS,RC/CSO,?PO,?O and ?EO were highest,and yet the light quality of G and Y treatment were quite the contrary.Indicated that RBY treatment promoted the PS? activity,boosted its light energy utilization.While G and Y treatment inhibited the photosynthetic characteristics.Above all,the nutritional quality were best and the light energy utilization were highest under RBY treatment.3.The quality and photosynthes of purple cabbage was affected significantly by different drought treatment.(1)The anthocyanin,flavonoid,vitamin C,soluble sugar,soluble protein content increased first and then decreased,and reached the greatest under T1 treatment;(2)Along with the increasing degree of drought treatment,the photosynthetic pigments and WUE increased slowly first and then decreased sharply,it illustrated that the appropriate water stress could improve the water use efficiency of purple cabbage.Pn,Tr,Gs all decreased,however Ci present the opposite tendency,showed that the decline of photosynthetic rate was caused by stomatal factors.The value of TRO/CS?DIO/CS?ABS/RC?TRO/RC?ETO/RC?DIO/RC?MO?VJ?WK all increased,and PIABS?VPO?ETO/CS?RC/CSO??O??EO all decreased with the increasing degree of drought treatment.Compared with CK treatment,the fluorescence data changed little under the T1 treatment.With the increasing degree of drought treatment,the electron flow from the side of PSII donor to the PSI receptor occured the significant changes in the fluorescence parameters,indicate that the drought treatment damages the system between the ? and ? photosynthetic electron transport chain,and maked the reaction center electron donor and the receptor electron all damage.Above all,the nutritional quality were best and the light energy utilization were highest under T1 treatment.4.The quality and photosynthes of purple cabbage was affected significantly by different chilling treatment.(1)Among with the temperature decreasing,the anthocyanin,flavonoid,soluble sugar,soluble protein and vitamin C content present rised first and fell later variation tendency.The anthocyanin and flavonoid content were higest under T? treatment and the soluble sugar,soluble protein and vitamin C content reached the greatest under T? treatment.(2)The photosynthetic pigment content and the value of Pn,Tr,Gs,PIABS,ETO/CS,RC/CSO,?PO,?O and ?EO all increased first and then decreased significant with the increasing degree of chilling treatment,and reached the greatest under T? treatment.However,the value of Ci TRO/CS,DIO/CS,ABS/RC,TRO/RC,ETO/RC,DIO/RC,VJ,WK and MO presented the opposite tendency.Illustrated that the appropriate chilling treatment could activate the PS? activity,and increased the utilization rate of light energy.But under the excessive low temperature condition,the blade electron transfer was restrained,and the activity state of PS ? reaction center was damaged,thus produced the adverse impact for the growth of purple cabbage.Above all,the nutritional quality were best and the light energy utilization were highest under T? treatment.
