Physiological fruit drop of Camellia Oleifera Abel is a major problem which influences the yield of Camellia Oleifera Abel.In order to illustrate the phenomenon of physiological fruit-drop in protein level,seeds dropped physiologically and seeds in the different stage of Min43 were collected in this research,the expression and function of differential protein base on the technologies of 2-DE and mass spectrometry in a level of protein of one breed of Camellia Olefera was analyzed.Last but not the least,this research aimed to suggest new solution for the problem of physiological dropped fruit.Moreover,a suitable 2-DE system for Camellia kernel was optimized,including sample protein extraction,IEF program,SDS-PAGE pouring,etc.The major results are as follows:(1)Conclusions made from the 2-DE system for Camellia Oleifera kernel proved that method of combing TCA/Acetone with SDS/phoel is better suitable for Camellia oleiferal kernel protein extraction.Also,sample in-gel rehydration as well as IEF program with low voltage for getting rid of salt ion and 62000vhr total focusing time was benefit for protein horizontal segregation.In vertical segregation,10%concentration of SDS-PAGE gel displayed a better result.(2)2-DE analysis of six different development periods of Camellia Oleifera kernels base on ratio>2 and anova<0.05 shows that:There are 41 proteins expression quantity changes compared with kernels between June and July.Compared with kernerl in July,kernel in August shows 158 protein variation.Compared with kernel in August,the kernel in September have 29 proteins are raised or lowered.compared with kernerl in September,the kernel in October have 1 protein expression quantity changed,compared with kernel in October,the kernel in November have 1 protein expression quantity changes.(3)2-DE coupling with MS,applied to study Camellia Oleifera Physiological fruit drop,showed that 17 protein spots from 21 detectable protein were successfully identified by MS.These proteins were categorized into the following six groups by function:cell growth,protein transpose and synthesis,energy metabolism,signal transduction,cell division and others.Furthermore,one differential protein identified as alpha tubulin 1 was lowly expressed in falling fruit,which relevant with the development of pollen,and low temperature stress inferring as a factor lead to the phenomenon of physiological fruit drop of Camellia Oleifera Abel.
