This study used the41Helianthus tuberosus L.germplasm resources thatcollected by Qinghai Academy of agriculture and Forestry Sciences to research bySRAP markers, based on the SRAP data and combining with morphologicalcharacter, analysis of H. tuberosus different germplasm resources of genetic diversity.The experimental results are as follows:1. To41of H. tuberosus resources2years botanical character survey statistics,mainly including the ground part and underground part of the character. The resultsshow that:41of H. tuberosus resource tuber character mainly include: fusiform,irregular nodular, rod, ball, wedge. Tubers color mainly has: purple, purple red,white, pink. The main characteristics of the part mainly includes: leaf color, leafshape, fluff, etc.2. Established the SRAP-PCR reaction system. The optimal PCR(20μL)mixturecontained2μL10×PCR Buffer,2.5mol/L Mg2+,0.24mmol/L dNTPs,0.25umol/L thepositive and negative primers,0.8U TaqDNA polymerase and80ng DNA template.22pairs of primer combinations with stable amplification and rich polymorphismselected from110pairs of primer combinations. Using screen out22pairs of primercombinations to41of H. tuberosus resources SRAP-PCR for amplification, the bandto the statistics, analysis, the establishment of0-1fingerprint.22pairs of primercombinations to41of material were amplification out160clear paths banding,including polymorphism banding article120(75%). SRAP amplification with grainis rich, amplification of DNA fragment size is in commonly between200to3000bp.3. Combined with41botany traits of H. tuberosus resources2years data toSRAP molecular genetic diversity analysis, genetic similarity matrix was establishedbased on160bands, the clustering analysis to construct kinship relationship tree,41of H. tuberosus resources distribution of genetic distance between0.65~0.95, withan average of0.80, the genetic similarity coefficient was0.72in cutting,41of H.tuberosus resources can be divided into two types: the first class I include Qingyu1and w16; the others are the second class II; the genetic similarity coefficient was0.80in cutting, the second class II can be divided into five subtypes: the first subtypeI include Qingyu2, w06, w18, w30, Qingyu3; the second subtype II include w09, w07, w24, w84, w64, w62, w66, s138, s150, w78, w79, w73, w69, w72and w32;the third subtype III include w23, w26, w22, w75, w36, w40and w43; the fourthsubtype IV include w50, w51, w61, w54, w19, w58, w02, w55, w59and w20; thefifth subtype V include w12and w39.
