Fatty liver hemorrhagic syndrome(FLHS)is a nutritional and metabolic disease characterized by lipid metabolism disorder,harmful to the development of layer breeding industry.The present study was designed to explore the role of lncRNA's abnormal expression in FLHS development process.300-day-old commercial Hy-Line layers were selected and randomly divided into two groups,with 20 replicates in each group.The control group was fed with basal diet,while the treated group was fed with high-energy-low-protein diet of diet.The whole experimental duration was 80d.Combined with H.E.staining and serum's TG and TC content,it illustrated that this experiment successfully built the FLHS model.3 liver samples were collected from control group and treated group respectively for RNA-Seq.The value for considering differential expression of transcripts was 2-fold difference.Thus,the genome-wide lncRNA and mRNA expression profiles were obtained.980 differentially expressed mRNAs were detected containing 475 down-regulated and 505 up-regulated ones,while 167 lncRNAs were differently expressed with 61 decreasing ones and 101 increasing ones.For the purpose of exploring the mRNA and lncRNA differences'impact on FLHS,their potential biological functions and the pathway they are involved were evaluated by utilizing GOseq method and KOBAS(2.0)software regarding GO and KEGG analysis.Differentially expressed mRNAs'GO enrichment includes cofactor binding,coenzyme binding,etc;Significantly enriched KEGG is the lysine degradation pathway.LncRNA may implement its biological function by regulating mRNAs.Cis and trans effect were utilized to predict target genes of differentially expressed lncRNAs in this study.Cis and trans effect of target genes were performed to GO and KEGG enrichment analysis respectively.Cis GO enrichment embraces NAD~+kinase activity,NADP biosynthetic process and cofactor synthesis,etc;Trans GO enrichment contains the nucleus,intracellular part and intracellular,etc.Cis KEGG enrichments are Notch signaling pathway and Fatty acid metabolism;Trans role KEGG enrichment is endocytosis pathway.Meanwhile,14 differently expressed genes regulating glucose and lipid metabolism were screened out.Regulator lncRNAs of this gene set were predicted according and trans effects.In order to verify the reliability of RNA-Seq data,these mRNAs and corresponding lncRNAs were selected for expression validation,with results consistent with the high-throughput sequencing data.This study obtained FLHS lncRNA and mRNA expression profiles,and some lncRNAs'abnormality is speculated to have close relationship with lipid disorder and oxidative stress in FLHS.
