Abnormal Expression Of Autophagy And Apoptosis-related Mrna In Liver, Kidney And Intestine Of Laying Hens With Flhs And Improvement Function Of Resveratrol

The purpose of present study was to investigate the changes of autophagy and autophagy-related mRNA expression in liver,kidney and intestine of Hy-line Brown laying hens with Fatty liver hemorrhagic syndrome induced by high-energy-low-protein diet.144 Hyline brown laying hens were randomly distributed into control group(Con),pathologic group(HELP),preventive group(HELP+Res)and resveratrol group(Con+Res)respectively with equal number.The experiment period was 120 days.The laying hens were sacrificed with an overdose intravenous injection of sodium pentobarbital(Nembutal,Abbot Labs,IL,USA;100 mg/kg)on days 40,80 and 120.The liver,kidney and intestine samples were collected from 12 Hyline brown laying hens in each group on days 40,80 and 120 respectively.Detected the expression level of ATG5、ATG7、Beclin-1、P62、Caspase-3、Bax、Bcl-2、HSP60、HSP70、HSP90、apoA-Ⅰand apoB-100 mRNA by real-time fluorescence quantitative PCR.The results were as follows:1.Compared with Con,the expressions of ATG5,ATG7 and Beclin-1 mRNA were decreased in HELP and increased in Con+Res in all tissues(P<0.01).Compared with HELP,ATG5,ATG7 and Beclin-1 mRNA expressions were increased in HELP+Res,the P62 m RNA expressions were contrary to them in all tissues(P<0.01).2.Compared with Con,the expressions of Caspase-3 and Bax mRNA were increased in HELP and decreased in Con+Res in all tissues(P<0.01).Compared with HELP,Caspase-3 and Bax mRNA expressions were decreased in HELP+Res,Bcl-2 mRNA expressions were contrary to them in all tissues(P<0.01).3.Compared with Con,the expressions of HSP60,HSP70 and HSP70 m RNA were increased in HELP(P<0.05)and no obviously change in Con+Res in all tissues(P<0.05).Compared with HELP,HSP60,HSP70 and HSP70 mRNA expressions were decreased in HELP+Res in all tissues(P<0.05).4.Compared with Con,the expressions of apoA-Ⅰ and apoB-100 mRNA were decreased in HELP and presented increased in Con+Res(P>0.05).Compared with HELP,apoA-Ⅰ and apoB-100 mRNA expressions were increased in HELP+Res in all tissues(P<0.01).In Conclusion,the expressions of autophagy and apolipoprotein mRNA were presented decreased,meanwhile,apoptosis and heat shock protein mRNA were presented increased in liver,kidney and intestine of Laying hens with FLHS induced by high-energy-low-protein diet.In addition,diet added resveratrol could increase the expression of autophagy and apolipoprotein m RNA,decrease the expression of apoptosis mRNA.
