In this experiment,the stem tip and top blade of tissue cultured seeding of Lycium ruthenicum were used to be experimental material and the colchicines as inducer,method adopted colchicines induced agent added to the medium,Besides,a preliminary method for identifying such polyploidy of Lycium ruthenicum was established by comparing with the diploid to acquire the polyploid plants of Lycium ruthenicum,which lays the foundation on the innovation,protection,and utilization of germplasm resources,the results were as follows:1.As the experimental material,the stem tip and top blade of tissue cultured seeding were treated by adopting induce agent added to the medium.Two treatment material in colchicines solution concentration was 500 mg·L-1,survival rate treated 7 days approach approximately median lethal dose.Therefore,the optimum treatment concentration of colchicines was 500 mg·L-1?treatment time was 7 Days.2.The stoma density,guard cell size and the number of guard cells of chloroplast,which was treated by blade materials were significant difference compared with the control,however,the majority of stem tip treated materials were significant difference compared with the control,the minority of them had no difference.Combined with chromosome observation,blade was more suitable as the material of induction of polyploidy.3.According to the blade of Lycium ruthenicum related to anatomy character and chromosome observation,preliminary screened out 7 tetraploid plant.Respectively,The top blade of Lycium ruthenicum act as treatment material,the combination of the concentration of colchicines was 300 mg·L-1,,treatment time was 7 Days;the combination of the concentration of colchicines was 300 mg·L-1,treatment time was 21 Days;the combination of the concentration of colchicines was 400 mg·L-1,,treatment time was 14 Days;the combination of the concentration of colchicines was 500 mg·L-1,treatment time was 7 Days.It was successful to induce tetraploid.The stem tip of Lycium ruthenicum act as treatment material,the combination of the concentration of colchicines was 4004.mg·L-1,treatment time was 7 Days;the combination of the concentration of colchicines was 500 mg·L-1,treatment time was 7 Days;The combination of the concentration of colchicines was 500 mg·L-1,treatment time was 14 Days.It was successful to induce tetraploid.
