In this paper,I used the combination of 11 Indica rice lines and 1 glutinous rice lines as test materials,to research the factors affecting anther culture of Indica rice,and to establish the system to cultivate Indica rice through the anther with high efficiency.The optimized Indica rice anther culture conditions are as follows.(1)Since when The distance between the base of the flag leaf and the auricle of the last leaf was 4-6cm,the main microspores stayed in the later uninucleated stage,the rice lines with 4-6cms' pulyinus distance are the best choices.(2)Genotypes of the Indica materials are an important factor restricting Indica anther inducing rate and directly affect the success and efficiency of Indica rice anther culture.(3)The different effects among culture mediums used in this experiment has not yet reached a significant high level,but the introduction rates of N6 or M8 medium were slightly higher than that in He5 medium(4)The induction medium that using 2,4-D 2mg/L + NAA 1mg/L + KT 1mg/L can significantly improve callus induction rate,which can be used as hormone combination of Indica rice anther culture.(5)To investigate the effect of temperature on Indica rice anther culture,I discovered that cultivation at high temperature at 31 ± 1 ? and at low temperature at 26 ± 1 ? showed no significant difference in the induction rates of anthers,but under conditions of 31 ± 1 ? callus grew significantly faster than the low temperature 26 ? 1 ?.(6)When using colchicine for Indica rice anther culture experiments,I found out:the difference between induction rate colchicine treated anther callus and untreated anther callus did not reach a significant high level,means colchicine did not increase the rate of anther callus induction.(7)When researching the sucrose's concentration on the impact of Indica rice anther culture,the results show:With the sucrose concentration increasing,the callus induction rate increases.60g/L of sucrose can improve the induction rate to the highest level,to be more specific:line 1316 reached 6.67 percent,and line 1317 reached 5.33 percent.When researching the callus' differentiation and rooting,we found:(1)The rice lines'genotypes are still an important factor of Indica rice lines' differentiation.Different Indica lines lead to differences in callus,and some Indica rice lines cannot produce green plants or may differentiate into albino.(2)Different concentrations of 6-BA has asignificant difference in improving the rate of indica rice anther differentiation.different concentrations of KT has not significantly increased the rate of indica rice anther differentiation.(3)Using sucrose or maltose had no huge difference in the rate of Indica rice anther callus' differentiation,but high concentrations of sucrose and maltose had an significant effect on the Indica differentiation rate.sucrose or maltose(60g/L)can reached the highest efficiency of differentiation.(4)In sum,rice line 688's differentiation rate using N6 medium is 4%,and rice line 1071's differentiation rate using N6 medium is 59%,rice line 1756's differentiation rate using MS medium is 11%,rice line 3073's differentiation rate using MS medium is 9%,rice line 3088's differentiation rate using MS medium is 4%.The number of other materials either undifferentiated or differentiated into albino plants,were not counted.The goal of this study is to establish a system for Indica anther cultivation and lay the solid foundation for future study.
