Horizontal gene transfer(HGT)is relative to the parent to offspring vertical gene transfer(VGT)and the proposed means kinship near or far between biological organisms are not carried out by the reproductive exchange of genetic material,or genetic information transfer.In this study,Cistanche deserticola and its host Haloxylon ammodendron as research material,Cistanche tubulosa,Cistanche salsa;Haloxylon persicum as a control,There are two molecular biology methods were using in this reasch:AFLP and transcriptome sequencing,and bioinformatics software such as DNAMAN,BLAST,Clustal X,MEGA6 were used to consider whether there is horizontal gene transfer between host Haloxylon ammodendron and parasite Cistanche deserticola,The results obtained are as follows:1.Through AFLP technology selective amplified by 117 consistent size and strip than a single capillary electrophoresis bands,and successfully recovered 43 which connect the carrier sequencing,sequencing success 31 of sequencing results were compared to obtain a sequence of 11 high consistency,proven:9 sequence of horizontal gene transfer does not take place,and the rest two for rRNA sequences,can not transfer its judgment whether the level of occurrence.2.Using RNA-seq sequencing five species such as Cistanchedeserticola,Cistanche tubulosa,Cistanche salsa,Haloxylon ammodendron and Tamarix chinensis transcriptome received a total 35176376880nt data.Assembly results:Unigene120797 a total length 108501802nt,the average length of 898nt,N50 reach 1845nt.Unigene functional annotation,respectively comments to NR 275000 个,NT 211943 个,Swiss-Prot 167204 个,KEGG 162922 个,COG 108443 个 library and GO 193 694,Unigene comment on all that is 292,117;predicted protein-coding frame(CDS)than for the protein library has 275,496 CDS,CDS has predicted 27,801,a total of 303,297;SSR total of 118,765.3.The results of this assembly transcriptome sequencing and shoots Cistanche consistency than right,received a total of 866 high-consensus sequences using Clustal X and MEGA6 software,using the maximum likelihood method,respectively,compared to the sequence and Blast work together to build a high consensus sequences obtained phylogenetic tree,a total of 131 mRNA sequences to identify suspected metastasis.The results will reveal Cistanche deserticola nuclear genes and host Haloxylon ammodendron horizontal gene transfer mechanism for exploring the evolution of inter-Cistanche their host Haloxylon provide a theoretical basis,and carry out the molecular genetic basis for breeding research foundation. |
