Lacrimal duct thrombus therapy is a common clinical treatment for dry eyes.According to its materials,it can be divided into permanent and degradable lacrimal duct thrombus.There are too many side effects of permanent lacrimal duct thrombus,and the time of degradable lacrimal duct thrombus staying in lacrimal duct is too short,so it is very important to develop new lacrimal duct thrombus.Corneal stromal lens(CSL)is a kind of non vascular tissue,which is widely welcomed by the field of corneal replacement.However,the application of CSL in implantable materials is still badly needed.Therefore,the preparation and application of CSL/SA/AM lacrimal duct suppository and it`s treatment in dry eye rabbits were studied;The main work of this paper is as follows:(1)The effect of monomer content on the properties of CSL/SA/AM lacrimal duct suppository was studied,and its preparation process and degradation performance were evaluated.When the content of SA is 27%(w/w),the gel network density is large,and the pore size is the smallest.Storage modulus 52.4Pa,tensile stress 0.10054 MPa,impact strength 16.787KJ/m2;heating up to 180?,the thermal weightlessness rate is 43.32%.The CSL/SA/AM lacrimal duct thrombus was prepared by the modified CSL.The surface of the tear duct thrombus was smooth and the size was uniform.The water absorption reaches 1.98 after 2 hours,which can effectively fit the lacrimal passage without extra pressure.The degradation time of degradable lacrimal duct plug was prolonged.(2)The dry eye model was made by 0.1% BAC,and CSL/SA/AM lacrimal duct plug was used to treat the dry eye after the model was finished,and compared with AA lacrimal duct plug,the main material of smartplug.The biological evaluation of lacrimal duct thrombus was carried out by the methods of lacrimal secretion,tear film rupture time,corneal fluorescein sodium,tiger red,richamine green staining,confocal microscopy,corneal SEM,TEM,H&E staining,protein imprint detection,etc.After the treatment of CSL/SA/AM lacrimal duct thrombus,compared with the model group,the amount of lacrimal secretion increased;the rupture time of tear film was prolonged;the scores of corneal fluorescein sodium,tiger red and ritain green staining decreased;the number of inflammatory infiltrating cells decreased;the surface of cornea was smooth,the number of corneal microvilli shortened increased;the layers of corneal epithelium were arranged orderly;the overexpression of NF-?B and P-NF-?B protein in corneal tissue was Inhibition.The results showed that CSL/SA/AM lacrimal duct thrombus was more effective than AA lacrimal duct thrombus in the treatment of dry eye.(3)the preparation technology of AG was studied,and AG/AM drug loaded gels were prepared,and then prepared into CSL/AG/AM drug loaded lacrimal duct embolus.The amino content in AG and the rheological properties,thermal properties,mechanical properties,drug release properties,CSL/AG/AM lacrimal passage structure and water swelling property of AG/AM drug loaded gels were tested.The results showed that when the mass ratio of gelatin to ethylenediamine was 10:1.4,the content of AG amino group was the highest.When L:AG=1:10 was used in AG/AM gel system,the pore size distribution of the gel system was uniform,the size was moderate,the modulus could reach 7000 Pa,the tensile stress reached 57 KPa,the gel impact strength was 14.31KJ/m2,the temperature weight loss rate was 55%,when heated to 80?.The drug release was 82.6% in 33 days.Therefore,L:AG=1:10 AG/AM hydrogel is suitable for modified CSL system.Prepare the lacrimal passage suppository.The surface of the drug loaded lacrimal duct plug is smooth and the size is uniform,and the maximum water swelling rate is 1.99 and the maximum diameter is about 1400?m after two hours of water soaking.(4)The model of glaucoma was made by injecting betamethasone/hydroxyethyl cellulose solution into the conjunctiva of the rabbit ball.After the model was made,CSL/AG/AM lacrimal duct plug was used for the treatment of glaucoma in the rabbit,and compared with the routine point drug method.IOP,retinal potential and H&E staining were used to evaluate the biological characteristics of the retina.The results showed that the IOP of CSL/AG/AM lacrimal duct thrombus decreased,the wave a and wave b of retina potential increased,the thickness of retina tissue increased,and the arrangement of various layers of tissue became more and more orderly.It shows that CSL/AG/AM lacrimal duct thrombus has excellent therapeutic effect on glaucoma.However,the therapeutic effect is slightly worse than that of conventional point drug therapy.
