Diabetes is a disorder of glucose metabolism caused by damaged islet B cells,insufficient insulin secretion or insulin resistance.Nowadays,the number of diabetics aged 20-79 years has reached 463 million,and the number of diabetics in China is116.4 million,ranking first in the world.In 2019,about 4.2 million people?aged20-79?died of diabetes or its complications,and the mortality rate accounted for11.3%of all deaths worldwide.At present,the clinical treatment of diabetes mainly depends on the injection of insulin,oral sulfonylurea,biguanide and other hypoglycemic drugs,but long-term use of hypoglycemic drugs will have greater toxic and side effects on human health.Rosa?roxburghii Tratt.?is a special resource in Guizhou.It is of extremely high nutritional value and good development prospects.This article aims to develop a natural plant product with obvious hypoglycemic effect by studying the hypoglycemic effect of Roxburgh rose and compounding it with some natural plants to bring the gospel to diabetics.The main research contents and conclusions are as follows:?1?Research on the hypoglycemic effect of roxburgh roseTo study the hypoglycemic effect of high?8 times concentrated?,medium?4times concentrated?and low?2 times concentrated?dose groups of roxburgh rose concentrated juice on streptozotocin?STZ?induced formation of type 1 diabetic mellitus mice.After successful modeling,the mice were randomly divided into model group,positive group,high dose juice,medium and low dose groups,and a blank control group was set up.After gavage,the body weight,water intake,fasting blood-glucose?FBG?,glycosylated serum protein?GSP?,glycosylated hemoglobin?GHb?,insulin?insulin?,INS))and other indicators were measured.The results showed that after 4 weeks of continuous intragastric treatment,the symptoms of polydipsia and polyphagia in diabetic mice was improved significantly,and the decline in body weight was suppressed.Compared with the model group,the FBG value and GSP of the high,medium and low dose groups of roxburgh rose juice were reduced to varying degrees,and the high-dose group had the best effect.It shows that roxburgh rose juice has an adjuvant treatment effect on type 1 diabetic mellitus mice,but considering the cost,the roxburgh rose juice is selected as the main raw material for the preparation of oral liquid.?2?Selecting research on formula accessoriesTartary buckwheat,balsam pear,mulberry leaf,propolis,and ginkgo biloba were selected as the raw materials for the initial screening.The fasting blood glucose?FBG?and fructosamine?FMN?in diabetic mice were used as the main reference indicators to determin the strongest hypoglycemic effect.The results showed that among the five raw materials,tartary buckwheat and bitter gourd had the best hypoglycemic effect,followed by ginkgo leaves and propolis,and mulberry leaves had the worst.Especially compared with the thorny pear juice group,the FBG values of the buckwheat group and the balsam pear group were reduced by 0.63%and 4.95%,respectively,and the FMN values were reduced by 11.08%and 0.52%,respectively,indicating that the buckwheat juice and balsam pear juice had better hypoglycemic effect than the roxburgh rose juice,so choose tartary buckwheat juice and bitter gourd juice as a recipe adjunct.?3?Design and optimization of oral liquid formulaUsing raw pear juice,buckwheat juice and bitter gourd juice as raw materials,the mixture design is used to determine the proportion of each component.Using the INS,GSP,and GHb values of diabetic mice as response values,the oral liquid formulations with no significant difference in the hypoglycemic efficacy during the three groups R1,R2,and R3.According to the sensory evaluation,the formula was rechecked.The R1 group had the highest sensory score of 76.08 points.The proportion was 57.34%for roxburgh rose juice,27.66%for tartary buckwheat juice,and 15.00%for bitter melon juice.The INS value was 35.17 m U.L-1,the GSP value was 2.18 mmol.L-1,and the GHb value was 147.23 ng.m L-1.Because of the compound oral liquid contains a lot of soluble solids,it is easy to precipitate after storage for a period of time,so stabilizer must be added.The optimal addition amount of stabilizer was determined by orthogonal test:xanthan gum was 0.06%,pectin was0.06%,cmc-na was 0.07%.?4?Research on storage stability of oral liquidThe stability of the oral solution was studied through light,high temperature,and accelerated experiments to determine the changes of the oral liquid's related properties,pH value,relative density,soluble solids,VC,total acid,and total flavonoid content under various environmental conditions.The results showed that:light has no obvious effect on the properties,pH value and relative density of roxburgh rose hypoglycemic oral liquid,but with the extension of light time,the content of VC and flavonoids had decreased.For better preservation solution,brown reagent bottle irrigation should be used Outfit.The temperature had a great influence on the properties,pH value and nutrient content of thorn pear hypoglycemic oral solution.With the extension of storage time,the color of the oral solution gradually changes from light yellow to light brown,with a slight precipitation,and the pH value gradually decreased.The content of nutrients gradually decreases,indicating that temperature and storage time havd a greater influence on the oral solution.Secondly,the microbial indicators in the oral solution after 3 months of storage were tested.As a result,the total number of colonies,E.coli,and molds did not exceed the standard,meeting national standards.?5?Evaluation of hypoglycemic efficacy of oral liquid and study on acute toxicityThrough in vivo experiments in mice,the hypoglycemic effect of the R1 oral solution after storage and the acute toxicity study were evaluated.The results showed that the stored R1 oral solution can effectively improve the dietary water consumption of diabetic mice,inhibit body mass decline,improve sugar tolerance,and effectively reduce FBG,GSP,and GHb values.Compared with before storage,the hypoglycemic effect of the oral solution in group R1 was reduced,but the hypoglycemic effect was better than that of single raw material tartary buckwheat juice,bitter gourd juice and Roxburgh rose juice.In the acute toxicity experiment,there was the death of the mouse.It was suspected that it was caused by improper operation in the midst of gavage,but the specific cause of death needs further research.
