Lung cancer is one of the malignant tumors with the fastest growth in morbidity and mortality,which is a series threat to human health.By early diagnosis,the disease can be detected as early as possible,which could effectively reducing the mortality.Compared with traditional screening methods of lung cancer,breath detection has the advantages of non-invasive,painless,convenient sampling and reasonable price,which is expected to be a new method of early detection,achieving early screening of lung cancer patients.This study intends to establish a standardized sampling procedure.The Pre-concentration-system and gas chromatography mass spectrometer(Pre GC-MS/FID)was used to analyze the breath VOCs of lung cancer patients,and108 kinds of VOCs were detected.The OVOCs samples were derivatized with2,4-Dinitrophenylhydrazine(DNPH),and then analyzed using ultra-high liquid chromatography-mass spectrometry(UPLC-MS/UV),by which 20 kinds of oxygenated VOCs(OVOCs)were detected.Through classification,the orthogonal partial least squares discriminant analysis(OPLS-DA)model was established to judge the difference of exhaled breath between sampled lung cancer patients and the healthy controls.The substances which have a greater contribution to the difference between the two groups of samples were screened out and listed as potential biomarkers.The screening characteristic VOCs included ethylene,2-butanone,methyl methacrylate,tetrahydrofuran,propane,1,4-dioxane,1-pentene,2-methylpentane,acetone,ethanol,butadiene,2,3-dimethylbutane,ethyl acetate,n-hexane,n-pentane,1-hexene,isobutane,2-methoxy Methylpropane,isopropanol and trans-2-butene.The characteristic OVOCs included formaldehyde,2-butanone,cyclohexanone and acetaldehyde.Furthermore,the screening characteristic VOCs and OVOCs were verified using logistic regression model,and it was found that the sensitivity and accuracies of the results were very high.In this study,a more comprehensive offline screening method was established for lung cancer patients,which provided preliminary reference and dataset forsubsequent study on breath VOCs biomarkers screening.
