In resent years bio-chip technique has developed to be focus in study of modern science gradually. The bio-chips invented presently include gene-chip, protein-chip, cell-biochip, electronic-biochip, tri-dimension-biochip, microflow-biochip, lab-on- chip, and chip, cDNA-chip, multi-peptide chip, spectrochip etc.Bio-chips above mentioned have been applicated in many areas of molecular biology, prevention, diagnosis and cure of disease, R&D of drug, monitor of environment and food safety. Especially in the field of drug R&D more and more people become interested in the new technique.In the experiment we monitor the hybridization of gene in real-time and in situation quantitatively ,by detecting the changes of frequency on the basis of Piezoelectric biosensor technique and study primarily the use of Piezoelectric biosensor technique in the screening of anti-lung cancer drugs.P53 gene is a cancer-inhibitor studied widely with the highest mutate-rate.It is valuable in predication of anti-drug ability and clinic study. So we use P53 as probes and 801-D lung-cancer cell lines of human as model of screening. After immobiling P53 on Piezoelectric biosensor we can detect signals of frequency during special gene fragment in solution hybridize with probes to characterize the gene and determine quantitative analysis. The experiment results show: (1)in liquid-phase conditions the Piezoelectric bio-chip analysis device is relevantly stable and the resonance frequency can vary in 3Hz.The auto-assemble and immbilization of probes can be finished in 1h. (2)according to the experiment results and Sauerbrey equation AF is simply relevant with Am when silicon wafer surge in proper vapor-phase and liquid-phase. Thus silicon wafer can be made into QCM with the detection limit of ng, even pg level, (3)when three drugs effect on the experiment we detect obviously changes of frequency (basis of piezoelectric biosensor) ,that is, the effection of drugs on P53.The result is the same with that of PCR method.In resent years the study of drug screening mainly focus on molecule level.Piezoelectric biosensor technique used in the thesis has made proper attempt Not only can Piezoelectric biosensor technique detect sample in real-time and in situ, but detect many samples in parellel. Although inexpensive cost, short time, no need of labeling, but it has many drawbacks. Protein-biochip and cell-biochip will become the trend in the future study because of their advantages. In a word bio-chip must attribute to the pharmaceutical research ,such as the found of drug targets, the research of drug activities, high throughout screening of drugs, study of toxicology and the study of drug genomes, etc.
