Flesh fingered citron is a plant of the group of Citrus.Flesh fingered citron essential oil is a volatile aromatic oil with a fresh smell and a slight floral aroma obtained by pressing and steam distillation from its peel.It has high application value and physiological activity value. This thesis mainly studies the essential oil of Flesh fingered citron fruit in Jinhua,Zhejiang(the most famous place for Flesh fingered citron production),analyzes its aroma composition, and uses C.elegans as a model to study in detail the nematocidal activity and The mechanism of action, and its effect on antibacterial,anti-inflammatory,whitening,etc.,attempts to expand the application fields of Flesh fingered citron essential oil in cosmetics, health products, antibacterial and anti-inflammatory drugs.First, the essential oil of Flesh fingered citron was extracted by steam distillation and analyzed by gas chromatography-mass spectrometry(GC-MS).The analysis results show that Flesh fingered citron essential oil contains more than 30 compounds, most of which are monoterpene compounds,of which d-limonene(43.39%),?-terpinene(18.57%),and ?-pinene(2.88%))And ?-pinene(2.69%)and other four compounds are the most abundant, and also contain a small amount of terpene oxides.Second, macroscopic methods such as contact killing were used to evaluate the nematocidal activity and mechanism through experiments such as acute stress, longevity, reproductive rate, hatchability, and behavior. The experimental results show that the Flesh fingered citron essential oil has good nematocidal activity against C.elegans,LD50 = 1.48 mg / ml,and the four main components in the essential oil have different effects on lethal nematodes. The result is that d-limonene It can be killed by contact killing, which leads to a reduction in its hatching rate. When the nematodes are cultured at 2mg / ml,the hatching rate is only 9%;?-terpinene is highly toxic and ?-pinene is a strong repellent. The spawning ability of nematodes is reduced. The total number of eggs laid by 2mg / ml treated nematodes is 202,which is far lower than the number of untreated essential oils.?-pinene is highly toxic and will interfere with the normal growth of nematodes and make the body.Organs such as the head and the head are inhibited. The amount of bending of the body treated by 2mg / ml is 6.3±0.26,and the number of head swings is 110±2.87.At the same time,it also reduces the anti-oxidant performance of the nematode. When the dose concentration is 1.5 mg/ml Flesh fingered citron essential oil was about 20 hours, the survival rate of nematodes dropped below 50%,and the life span of survival was also reduced by 8 days compared with the control group.Thirdly, the microcapsules are embedded by the embedding method, and ?-cyclodextrin is used as the wall material to embed the Flesh fingered citron essential oil.Scanning electron microscopy and thermogravimetry were used for characterization. The optimal core-wall ratio of the microcapsule was 1: 6,the embedding yield was 96.24%,and the loaded capacity was 32.67±1.16%.At the same time, microcapsules and four major compounds were investigated for nematocidal tests and acute validation tests. The experimental results show that the stability experiment proves that the microcapsules have a sustained release effect, can slowly release the active ingredients, and have a long-lasting nematocidal effect. At the 4th hour, the lethal rate to nematodes reaches 77%.The lethal experiment of the compounds proved that within 24 hours, the lethality of d-limonene to C.elegans was 69.67%,?-terpinene was 66%,?-pinene was 88%,and ?-pinene was 90%.The results of the acute stress test further proved that the essential oils and the four major compounds could have a certain nematocidal effect by reducing the antioxidant capacity of C.elegans after being embedded in microcapsules.Fourth,Flesh fingered citron essential oil not only has a natural and unique aroma, but also has a bactericidal effect. Therefore, microcapsules were used as raw materials in this study.First, the antibacterial effect on Staphylococcus aureus and E.coli was verified. The experimental results showed significant inhibition.Bacterial circle. It also explored the patch test of Flesh fingered citron essential oil and the whitening experiment of inhibiting tyrosinase.Volunteer human patch tests have shown that within the range of 0-2% safe use,Flesh fingered citron essential oil has no irritation to human skin.At the same time, the results of whitening experiments that inhibited tyrosinase showed that the amount of Flesh fingered citron essential oil was as low as 0.5 mg/ml,and the final inhibition rate could reach 9.26%,and the inhibition rate range was 0.5%-9.26%.Starting at 20 mg/ml,it also has an inhibitory effect on tyrosinase, and the final inhibition rate can reach 16.3%.It proves that Flesh fingered citron essential oil has whitening effect. Therefore,a series of daily necessities were made by using Flesh fingered citron essential oil and its microcapsules, and products such as toothpaste, washing powder, shampoo, shower gel, body lotion, and soap were developed. The research results of this subject show that Flesh fingered citron essential oil and microcapsules have good nematocidal activity and daily application, and it is expected to be used to replace chemical synthetic pesticides and will have wide application value. At the same time, the results also help the development of new products and uses of Flesh fingered citron essential oils and fragrances, expand new markets, and give full play to the advantages of Flesh fingered citron essential oils.At the same time, they also achieve the purpose of promoting the development and application of citrus fruit and further promote the further processing of Flesh fingered citron plants industry development and expansion of its application fields.
