Tea and its main functional components(EGCG,theanine and caffeine)has been studied extensively for its beneficial health effects on anti-aging in model organisms such as Caenorhabditis elegans,Drosophila flies and mice.Based on previous studies,this study explored the effects of GA and EGC on the lifespan in Caenorhabditis elegans.And the interaction effects of combinations of EGCG,caffeine and theanine on the lifespan in Caenorhabditis elegans.The main research results are as follows:(1)Under normal culture conditions,the survival curve of wild-type N2 exposed to EGC or GA from adult indicated that 0-1000μmol·L-1EGC or GA had no significant effect on average life.Under oxidative stress and ultraviolet stress experiments of paraquat showed that200 and 1000μmol·L-1EGC and GA had no significant effect on the stress resistance of wild type Caenorhabditis elegans.(2)Under normal culture conditions,the survival curve of wild-type C.elegans N2 was observed.EGCG intervened from adult worms,and its average life span was significantly prolonged at 200μmol·L-1,while the average lifespan was significantly shortened at 1000μmol·L-1.Caffeine intervened from larval stage,and its average life span was significantly prolonged at 5-10 mmol·L-1,while the average lifespan was significantly shortened at 20mmol·L-1.0-1000μmol·L-1 theanine had no significant influences on mean lifespan.Under normal culture conditions,EGCG,caffeine and theanine(4:2:1)combined intervened wild-type C.elegans N2.The survival curve found that Compared with EGCG alone intervention group,200μmol·L-1 combined group has no significant difference in its average life span;1000μmol·L-1 combined group has a tendency to recover to its normal state.Under normal culture conditions,caffeine and EGCG combined with the effective intervention dose and time to intervened wild-type C.elegans N2.The survival curve found that the combinations of EGCG and caffeine at effective intervention dose and time had no synergy effect on the mean lifespan.(3)EGCG and theanine were combined to intervene lifespan in Caenorhabditis elegans.Compared with relatively EGCG alone intervention group,the mean lifespan was not further extended at 200μmol·L-1combinations.But the mean lifespan of combinations showed a tendency to recover to its normal state at 1000μmol·L-11 combinations.And theanine can effectively improve the body movement ability of C.elegans at relatively high doses of EGCG.The above experimental results showed that in the Caenorhabditis elegans model,the anti-aging activities of EGC and GA were weak in the nematode model EGC and GA.The combination of caffeine and EGCG has no significant synergistic effect on antiaging.Theanine can effectively alleviate the body’s recession response caused by high dose EGCG.
