Seabuckthorn is a kind of herbaceous plant rich in many kinds of active substances.Seabuckthorn seed protein?SSP?has significant hypoglycemic activity.The hypoglycemic activity of seabuckthorn seed protein enzymatic peptide?SSPE?and its effect on diabetic renal function have not been reported.Therefore,in this project,SSP was enzymolysis to prepare active peptide,and further explored its antioxidant,hypoglycemic activity and the impact on diabetic renal function.Under the experimental conditions in this paper:?1?The optimum conditions of pancreatin hydrolysis were,0.35%of enzyme volume,6 h of enzymolysis time and 11 p H.Under these conditions,the recovery rate and hydrolysis degree of SSP were 75.38%and 13.55%,respectively.There was no significant change in the percentage of amino acids between SSP and SSPE,but the total amount of amino acids decreased by 14.85%compared with that of SSP.The molecular weight of SSPE was significantly smaller than that of SSP as well.After enzymatic hydrolysis,the proportion of peptides with molecular weight<5 k Da increased,the proportion of peptides with molecular weight between 5-10 k Da changed little,and the proportion of peptides with molecular weight>10 k Da decreased.?2?The antioxidative effects of different concentrations of SSPE in vitro were studied.The results showed that the total anti-oxidation effect of SSPE was better than that of SSP,and it had strong reducing ability,scavenging ability of superoxide anion free radical and hydroxyl free radical,anti-lipid peroxidation ability.In addition,the anti-oxidation effect was proportional to the concentration,all at the highest concentration?1 mg/m L?showed the strongest antioxidant capacity.But the scavenging capacity of DPPH free radicals was weak,and the scavenging capacity decreased with the increase of concentration,with the maximum scavenging efficiency of 62.55%at 0.2 mg/ml.The inhibition rate of malondialdehyde in liver and kidney of mice induced by Fe2+-H2O2increased first and then decreased,and reached the maximum inhibition rate at 0.8 mg/ml.?3?The hypoglycemic activity of SSPE and its effect on diabetic renal function were discussed.The results indicated that SSPE?50,150,250 mg/kg/d?showed outstanding hypoglycemic activity and renal protective effect,and the hypoglycemic index was better than that of SSP.The glomerular filtration rate?the increasing of urine protein,creatinine,uric acid,urea nitrogen,malondialdehyde?,glomerular hypertrophy,basement membrane thickening,renal vesicle atrophy and other symptoms caused by early stage of diabetic nephropathy were also significantly improved.The best protective effect on kidney is the SSPE low dose group?50 mg/kg/d?.
