| In accidents such as traffic accidents,uncontrolled bleeding is an important cause of death.Hemostasis materials can quickly stop bleeding and reduce blood loss.It is of great significance to save people’s lives to win time for rescue arrival.Silk fibroin is a biocompatible and degradable natural polymer,and it also has a certain hemostatic activity.It has great potential for application in the field of hemostasis.This paper studied a silk fibroin gel hemostatic material,and studied its material properties,swelling mechanism and hemostatic propertiesIn this paper,silk fibroin was used as the substrate,and an initiator was added to the silk fibroin solution to promote its gelation to obtain silk fibroin hydrogel for hemostasis.Riboflavin is used to form free radicals under UV light and catalyzed by horseradish catalase(HRP)to promote tyrosine cross-linking in silk fibroin.On this basis,the degree of cross-linking of silk fibroin molecules was adjusted to obtain a silk fibroin hemostatic material which maintains an amorphous structure and has high swelling capacity.The results show that the silk fibroin hemostatic material prepared at a silk fibroin concentration of 20 mg/mL and a riboflavin concentration of 0.1 mM can adsorb 91 times the weight of deionized water.The silk fibroin hemostatic material has the ability to quickly absorb water in the blood and increase the viscosity of the blood,thereby increasing the concentration of the hemostatic active substance.Silk fibroin can form a gel to block the blood vessels of the wound after swelling with water.The swollen hemostatic material can be degraded within 5 days,thereby avoiding secondary injury caused by material removal.The hemostatic effect of silk fibroin material was tested,and its coagulation mechanism was explored.The results showed that the hemostatic time of silk fibroin hemostatic material was 48 s in vitro and that of rabbit ears was 68 s.The hemostatic effect was significantly better than that of medical hemostatic gauze.Silk fibroin hemostatic material accumulates red blood cells to increase blood viscosity,slow down the flow rate and promote hemostasis;its own negative electricity is favorable for platelet adhesion to activate endogenous coagulation;in addition,the adsorption of protein substances by silk fibrostatic hemostatic material can also increase blood viscosity and accelerate hemostasis. |
