Bleeding is the main cause of intraoperative and postoperative morbidity and mortality.Currently,hemostatic agents in clinical application are mostly based on natural materials and enhance hemostatic effect by forming coagulation structure.Hemostatic sponge made from natural polysaccharides has been paid much attention by researchers because of its good biocompatibility,high absorption rate and rapid blood cell recruitment.However,without the modification of polysaccharides,they are still having trouble in mechanical strength,fluid absorption rate and antimicrobial properties,so the polysaccharides were needed to be modified in order to make its have good biocompatibility,joint wounds,efficient fluid absorption rate,rapid raising effect of blood cells,appropriate antibacterial effect,no secondary wound infection,excellent mechanical strength and mild preparation conditions,etc.In this research,chitosan quaternary ammonium salt was prepared by modifying chitosan,which made it have good antibacterial and water solubility.Subsequently,caffeic acid was grafted onto chitosan quaternary ammonium salt to enhance the tissue adhesion of the material and achieve rapid hemostasis effect.In terms of the selection of crosslinking agent,pullulan polysaccharides were prepared as oxidized Pullulan polysaccharides with dialdehyde structure.Hydrogel materials were prepared by schiff base reaction with caffeic acid modified chitosan quaternary ammonium salt,and sponges with good antibacterial and hemostatic properties were obtained after freezedrying.The results show that the material has high absorption rate,good hydrophilicity,mechanical properties and antibacterial properties.The mechanical strength and biocompatibility of hemostatic sponge can be enhanced by compounding self-extracting silk fibroin in the system.The results showed that the silk fibroin glue was successfully extracted with lithium bromide and compounded into hemostatic sponge.The antibacterial and mechanical properties of sponge were greatly improved.Silk fibroin is a natural source and has no effect on the biocompatibility of sponge materials.Finally,the simulation of wound healing experiment was carried out.A wound model with a diameter of 1 cm was established on the back of SD rats,and the two hemostatic sponge materials were applied to the wound surface.The effect of the materials on wound healing was investigated by the appearance,healing rate,healing time and HE staining section of the wound.The experimental results showed that the hemostatic sponge with silk fibroin protein compound had better effect on wound hemostasis and promoting wound healing.
