| Monacolin K,the main active ingredient in functional red kojic rice,has the function of reducing blood lipid,thus having the function of medical care.So it has broad market.This paper first uses,a method for the determination of Monacolin K content in Monascus rice by using high performance liquid chromatography(HPLC).The peak area value of Monacolin K was obtained after detecting standard solutions with different concentrations.The regression equation was calculated as y=51.349x+0.1784,correla tion coefficient r2=0.9997.The peak time of Monascus Monacolin K in liquid chromatography is about 15 minutes.The recovery rate experiment and precision experiment were carried out on the method,and the lowest recovery rate was 97.42%,the highest recovery rate was 99.96%,the average recovery rate was 99.22%,and the recovery rate was above 98%.It is proved that the recovery rate is good.The relative standard deviation of precision is 0.028(<0.05),and the precision was good.After determining the detection method of monacolin K,we based on the above research,taking monacolin K content as an index,obtained 6 functional monascus strains from Shandong zhonghui food co.,ltd.and screened out one strain with the highest content,with monacolin k content of 10.48 mg/g.The strain was subjected to single factor experiments such as bran content,carbon and nitrogen sources,inoculation temperature,fermentation time and pH.After orthogonal optimization,the optimal medium composition of functional monascus rice with high yield of monacolin K was obtained:initial water content 50%,rice 50%,bran 10%,peptone 1.5%,glucose 2.5%.The optimal culture conditions were obtained:initial pH=5,inoculation amount 12%,rice particle size 14 mesh,bottling amount 60g,culture at 30℃ for the first 3 days,and low temperature fermentation at 26℃ for the next 15 days.Under the best conditions,the yield of monacolin K was 13.21 mg/g.The produced monascus rice powder is mixed with coenzyme Q10,spirulina and microcrystalline cellulose according to the ratio of 43:4:30:23 respectively to prepare monascus capsules.The acute oral experiment of mice was carried out with monascus capsule powder.After 14 days of observation,there was no obvious change in the health status and body weight of mice.Therefore,it was judged that monascus capsule powder was nontoxic.The content of Monacolin K in Monascus capsule powder after being digested by artificial gastric juice and artificial intestinal fluid is detected,and the content of Monacolin K is unchanged,with an average value of 3.47 mg/g,that is,the content of Monacolin K,the effective component,will not be reduced after being digested by artificial gastric juice and artificial intestinal fluid. |
