Persimmon leaves are the fresh or dried leaves of plant persimmon(Diospyros kaki L.)Persimmon leaves have a variety of effective compounds,containing a high nutritional value,with exert hypoglycemic and lipid-lowering,hemostasis,anti-hypertensive and anti-tumor and many other functions.The mechanism of hyperlipidemia and diabetic on animal model or human body is not fully understood.In this study,the model mice were fed with instant tea from persimmon leaves.The model animals were used to test the effects of their physiological parameters and the differential expression sequences of their liver transcriptomes.The effects of persimmon leaf instant tea on glycolipid metabolism and the main experimental process were preliminarily studied.The results are as follows.For hypoglycemic animal experiment(the experimental mouse is Kunming mouse),the purchased mice were divided into a normal diet group(Nd)and high carbohydrate model group(Hc),a high carbohydrate model fed with persimmon instant tea group(Hc-Pt),a high carbohydrate model administered with positive drug group(Hc-Pd).The mice in the Hc,Hc-Pt and Hc-Pd groups were induced to establish a high glucose model by high-carbohydrate diet combined with intraperitoneal injection of streptozotocin(STZ).Hc-Pt and Hc-Pd mice were treated continuously for 5 weeks.The results showed that,compared with the high carbohydrate model,Instant tea from persimmon leaves significantly reduced the liver weight and the content of serum low density lipoprotein cholesterol(LDL-C),total cholesterol(TC).It also the significantly reduced liver TC content,increased superoxide dismutase(SOD)content,and significantly reduced the concentration of alanine aminotransferase(GPT)and aspartate aminotransferase(AST)in liver injury indicators.This results indicates that persimmon leaf instant tea can enhance the antioxidant ability and alleviate liver damage in high glucose mice.The mouse liver transcriptome was sequenced,and the results of sequence comparison,functional annotation,functional enrichment and expression differences showed that,in the aspect of genes and signaling pathways related to glucose and lipid metabolism,persimmon leaf instant tea may significantly affect the metabolic pathways involved in the regulation of glycolipid metabolism,and play a role in the expression of some key genes in glycolipid metabolism.For example,persimmon leaf instant tea can significantly up-regulate the expression of growth factor receptor-binding protein 10(Grb10),and significantly down-regulated the expression levels of lipoprotein lipase(LPL),uncoupling protein 2(Ucp2)and fatty acid synthase(Fasn).For lipid-lowering animal experiment(C57BL/6J mice),the purchased mice were randomly selected 9 as the normal diet group(Nd').The other mice were induced by high-fat diet to establish high-fat model.The successful mice were randomly divided into high-fat model group(Hf),high fat model fed persimmon leaf instant tea(Hf-Pt),and hyperlipidemia model administered with positive drug group(Hf-Pd).Hf-Pt and Hf-Pd mice were treated continuously for 6 weeks.The results showed that compared with the high-fat model mice,persimmon instant tea significantly increase liver lipid SOD concentration.The mouse liver transcriptome was sequenced and analyzed by sequence alignment,functional annotation,functional enrichment and expression differences.The results showed that the persimmon leaf instant tea had a significant effect on the expression of liver lipid metabolism related genes in mice.And persimmon leaf instant tea could significantly affect the expression of acetyl coA carboxylase(Acaca),acylCoA thioesterase 3(Acot3),main urinary protein 17(Mup17),salt-induced kinase 1(Sikl)gene and regulate lipid metabolism levels.The results suggested that persimmon leaf instant tea has some positive effects on the metabolism of sugar and lipid in animal body,but those effects are very limited.In addition,we need to further study on the complex components of persimmon instant tea,which molecules play a role in the corresponding target gene.
