The rapid development of nanotechnology has brought about a new technological revolution.Many advantages of nanoformulations make it more and more popular,and its research and application in various fields are paid more attention.Based on the medical value of medicinal plants,it can not only play a great role in medicine,but also get a broader development in agricultural fields such as veterinary drugs and pesticides.The purpose of this study is to further develop and utilize medicinal plants to prepare new nanoformulations using nanotechnology.In this study,two kinds of nano formulations were prepared by processing medicinal plants with nanotechnology.Nanoparticle formulation is to crush the medicinal plants directly into nanometer powder.The dissolution behavior of its active ingredients was studied.Double emulsion nanoemulsion adjuvant was that the active components in medicinal plants were modified to prepare nanoemulsion adjuvant of medicinal plants using nanoemulsion carriers.The particle size,pH value,viscosity,stability and safety were measured,and the immune effects of the two nano adjuvants were evaluated by animal experiments.The contents were listed as follows:1.Panax notoginseng and Panax quinquefolium nanoparticles were prepared using wet ball milling.It was determined that the medicinal plants nanoparticles were prepared with a solid-liquid ratio of3.125%,a rotating speed of 2000 r·min-1,and grinding for 2 h.The nanometer powder of Panax notoginseng and Panax quinquefolium was prepared with particles size of 324.17±0.63 nm and 298.97±0.32 nm,respectively,with good dispersibility.The dissolution rate of ginsenoside in both nanometer powder was higher than that of ultrafine powder.The ginsenoside Re in the nanometer powder of Panax quinquefolium was the best,43.89%higher than that of the ultrafine powder.2.The medicinal plants emulsion nanoemulsion adjuvant was screened and evaluated.In this research,the particle size of the nanoemulsion were used as indicators to screen surfactants.It was found that when surfactants 2 and 3 were used in combination,and the ratio of surfactants 2 and 3 was 3:1 in the pseudo-ternary phase diagram,the area of the nanoemulsion was the largest,and the particle size was the smallest.Medicinal plants emulsion nanoemulsion adjuvant and blank emulsion nanoemulsion adjuvant were prepared using two-step method,two emulsions were clear and transparent with stable properties,water-oil-water nanoemulsion was identified using centrifugation and staining meathods,particle size of blank emulsion nanoemulsion adjuvant was 128.77±0.61 nm,PDI was 0.16±0.01,pH value was 5.81±0.33,viscosity was 3.8±0.15 s,particle size of medicinal plants emulsion nanoemulsion adjuvant was 139.21±0.44 nm,PDI was 0.13±0.32,pH value was 5.78±0.64,viscosity was 3.7±0.83s,and the stability were good.In the safety test,no adverse reactions were found in all the experimental animals,and all the animals were healthy.3.Medicinal plants of nanoemulsion adjuvant was preparated and its immune enhancement effect was verified by experiments.In this research,polysaccharide from Ganoderma lucidum was prepared into the medicinal plants nanoemulsion adjuvant.The emulsion was translucent and stable,particle size was143.03±0.72 nm,PDI was 0.09±0.02,pH value was 5.88±0.12,viscosity was 4.0±0.37 s,and the stability were good.The immune effect of mice was evaluated with two nanoemulsion adjuvants together with inactivated antigen.In the early stage of immunity,the two adjuvants had better immune enhancement effects than the other two adjuvants,and the medicinal plants nanoemulsion adjuvant was superior to nanoemulsion.In the later stage of immunization,the antibody titer of the two adjuvants increased slowly compared with 206 adjuvant,but the immune effect was better than the commercial adjuvant,which indicatied that both of the two nanoemulsion adjuvants could enhance the immune effect of the vaccine.
