Recently,The incidence of gout is rising significantly and become more and more popular in young,which has become one of the common metabolic diseases that plague human health.Diseases such as cardiovascular and cerebrovascular diseases,hypertension,diabetes and other diseases,and even lead to kidney failure are often caused by gout besides crystal arthritis.Western medicine for clinical treatment of gout is often accompanied by fever,rash,and serious liver and kidney The burden is strong,and it is easy to repeat.The active ingredients of medicinal plants are safer and have a wide range of pharmacological effects,which provides more choices for the development of functional foods for prevention and relief of gout.In this study,7 medicinal plants lotus leaf,ginkgo leaf,polygonum cuspidatum,radix paeoniae rubra,paeonia suffruticosa,radix puerariae and geranium were used for test.The content of active ingredient of all extracts were determined.With the Uric acid metabolase as the target,the biological activity of small molecule compounds was simulated and evaluated by molecular docking technology,and to evaluate the effect of reducing the uric acid.The content of flavonoids,polyphenols and polysaccharides of all extracts were determined.The extract of radix puerariae water extract was the highest,reaching45.24% as it contains more water-soluble polysaccharides.The highest total flavonoid content was Polygonum cuspidatum alcohol extract,which was 3.55%.The highest total phenolic content was ginkgo leaf alcohol extract,which was 6.51%.Ultraviolet spectrophotometry was used to determine the XOD inhibitory activity of plant functional components.Alcohol extract of Polygonum Cuspidatum with the IC50 value 2.366 mg/ mL which was the highest among all extracts.The relationship between the content of active substances and the inhibitory activity of xanthine oxidase was analyzed.It was found that there was no correlation between the polysaccharides content and the inhibitory activity of XOD,while the total flavonoids content and the inhibitory activity of xanthine oxidase was linearly correlated.Molecular docking simulations were used to filter the active compounds which have the effect of reducing uric acid by docking the small molecular compounds derived from five medicial plants and xanthine oxidase(XOD),adenine nucleosidedeaminase(ADA)and purine nucleoside phosphorylase(PNP).And comparing with TEI,FRK and IM5,the docking energy with XOD,ADA and PNP of 48,18 and 10 small molecules were quite low respectively.Among them,30.35% and 26.78% of XOD inhibitors were derived from polygonum cuspidatum and ginkgo biloba.ADA inhibitor lotus leaf contributed the most components(35.00%),then polygonum cuspidatum was 30.00% and ginkgo biloba was 25%.The contribution of PNP inhibitor polygonum cuspidatum was 35.71%,which was consistent with the results of in vitro xanthine oxidase inhibition experiment.The synergistic effect of 3,4,3',5'-Tetrahydroxystilbene-3-glucoside,Cianidanol,luteolin-7-o-glucoside,(+)-Epicatechin,Cosmetin,Cianidanol,(-)-catechin and Polydatin on XOD,ADA and PNP shows that the effect of medicinal plant reducing uric acid level had multiple targets.The determination of active ingredient content,in vitro uric acid lowering experiment and molecular docking,which provides strong support for the further extraction and separation of active substances,in order to make better use of medicinal plants,for the development of gout prevention Functional food lays the foundation.
