Study On Preparation Technology And Quality Standard Of Fangcang Granules

Objective:The best medicinal materials for Saposhnikovia divaricata were selected from seeds base source and made into fangcang granules.To study the preparation technology and quality standard of fangcang granules further.Methods:The seeds were tested by the quartering method for weight of a thousand seeds.Tetrazolium staining was used to determine seed viability.The paper bed was used to determine the germination rate of seeds by using the filter paper method,the reverse filter paper method and the absorbent cotton method.Combined with sem to screen better seeds.HPLC method was used to determine the content of Prim-O-glucosylcimifugin and4'-O-glucosyl-5-O-methylvisamminol in Saposhnikovia divaricata.Alcohol soluble and water soluble components were extracted by ethanol reflux and decoction.The single factor experiment and orthogonal experiment L9(3~3)were used to select the process parameters.The optimum extraction conditions were determined based on the yield of dry paste.Determine the best molding process according to the amount of auxiliary materials.TLC was used to qualitatively identify medicinal materials in granules.High performance liquid chromatography was used to determine the content of Saposhnikovia divaricata components in the granules.Results:From the thousand-grain weight:FF7>FF4>FF8>FF5>FF6>FF2>FF9>FF1>FF11>FF3>FF10;At each concentration,the living force was FF7>FF9>FF4>FF8>FF5>FF10>FF2>FF3>FF1>FF11>FF6;According to the germination experiment,the total germination rate of FF1,FF5,FF9 and FF2 seeds was higher than that of other seven seeds in the same germination days;By taking the content as the index,it can be known that the order of content is FF9>FF2>FF5>FF4>FF10>FF3>FF6>FF11>FF7>FF13>FF8>FF1;There were significant differences in sem results.Extraction technology of fangcang granules:The alcohol extract was added with 18 times 50%ethanol,the extraction time was 3h,and extraction 2 times.After alcohol extraction,the drug residue was soaked in 16 times of water for 0.5 hours,then boiled for 3h,and then extracted for 5times.Molding process:The auxiliary materials are soluble starch and dry paste powder:auxiliary materials(1:1)and 60%ethanol are used to make soft materials.Quality standard:TLC method was uesd to identify Saposhnikovia divaricata,Atractylodis rhizoma,Taraxacum mongolicum,Clematis chinensis osbeck,Stigma Maydis,Phellodendri,Chinensiscortex,Pheretima,Plantago,Prunus persica.HPLC method was used to determine the content of Prim-O-glucosylcimifugin and 4'-O-glucosyl-5-O-methylvisamminol in fangcang granules.Conclusion:The best extraction process and forming process of fangcang granules were determined.And the quality standard of fangcang granules was established,which provides a basis for improving its development and application.
