Porcine are one of the biggest source of global meat production and produce large amount of by-products such as hairs during processing.A massive amount of porcine hairs(a protein enrich product)is wasted.Hairs are difficult to degrade,and improper treatment can cause environmental pollution.Therefore,extraction of bioactive peptides from hairs is a way to recycle this waste.In this paper,we hydrolyze porcine hair keratin to prepared antioxidant peptides.The main research contents are as follows:1.The basic composition of porcine hair was analyzed.The hairs contain 90.89%protein,and 36.48%hydrophobic amino acids.The results indicate that porcine hairs are expected to be an excellent protein source of antioxidant peptides.2.Porcine hairs were hydrolyzed by alkali-reduction method and the scavenging rate of hydroxyl radical of hydrolysate was determined.The optimize conditions for the preparation of porcine hair keratin-based antioxidant peptide was determined by response surface optimization method:sodium hydroxide 6.9 g/L,hydrolysis time was 6.7 h,anhydrous sodium sulfite was 19.6 g/L,and the reaction temperature was 89.3 0C.3.The effects of external conditions such as pH and temperature on the stability of porcine hair keratin-based antioxidant peptides were investigated.The results showed that the effects of pH,temperature on the stability of antioxidant peptides were not significant.With the increase of salt concentration,the hydroxyl radical scavenging rate of antioxidant peptides was greatly reduced.After simulated gastrointestinal digestion,the ABTS and hydroxyl radical scavenging of antioxidant peptides increased.4.Based on the isoradiometric analysis and cell experiments,the biological activity of the porcine hair keratin-based antioxidant peptide was explored.The experimental results show that when the collagen peptide and the antioxidant peptide are mixed at a mass ratio of 4:1 and 9:1,the scavenging of ABTS and hydroxyl radicals and the growth of mouse fibroblasts synergistically increased.This indicates that the as-prepared porcine hair keratin-based antioxidant peptide has good biomedical potential.
