Studies have demonstrated that the soy protein have hypocholesterolemic activity,but the mechanism is not fully clear.In this paper,two in vitro models for micelles transport and fluorescent probe were employed to verify the hypocholesterolaemic activity of tofu peptic hydrolyzate.Lactone tofu and hard tofu,which had been shown to hold hypocholesterolaemic activity,were used as the research object.In vitro gastrointestinal digestion simulation model was applied to obtain different molecular weight(M_R)digestion fraction products.Detected by both HPLC-ELSD and detection kit,basing on the influence of products’cholesterol micelle on transportation,the two models,namely micelles transport and fluorescent probe were explored for determinaion.The gel chromatography and dynamic light scattering(DLS)were used to analyze the composition and distribution of microparticles in the artificial micellar model and observe the effect of inhibiting cholesterol transport by adding different concentrations of samples.The results confirmed that tofu hydrolyzate exerting hypocholesterolaemic activity,and different M_R fractios showing different inhibiting effects.The results also showed that soyprotein peptic hydrolyzates>10k HHT had the highest activity of cholesterol transport,and the absorbance of cholesterol was(55.34±0.03)%.There are two kinds of microparticle-phospholipid vesicles and mixed micelles occurrng in the artificial micellar model,and the two kinds of particles are involved in the transport of micelles in different degrees,and have different inhibitory effects at different sample concentrations.The fluorescence probe(DChol)has been successfully synthesized for the fluorescent probe model and the purity is larger than 90%,the hypocholesterolaemic activity of tofu hydrolyzate is further verified by the intuitive fluorescence.In conclusion,both micellar transport and fluorescence probes can be used to analyze the inhibitory effects of tofu hydrolyzate on cholesterol transport and absorption.
