To investigate the effect of SSW on intestinal flora in IBS-D rats.To explore the efficacy of this kind of prescriptions in improving IBS-D,and its relationship with intestinal flora and the rationality of compatibility with SSW.45 SPF male SD rats were randomly divided into normal group,model group,Positive control group,Sishen Wan group,Ershen Wan group and Wuweizasan group.The spleen-kidney-yang deficiency type IBS-D rat model was prepared by a composite factor and administered for 14 days.After collecting the feces of the rats,the total DNA of the stool sample was extracted.Primers were designed based on the 16 S r RNA V3 to V4 regions of the bacteria,and then subjected to high-throughput sequencing using the Illumina Miseq platform.At the same time,the content of marker proteins in the intestinal mucosa of rats and the concentration of related cytokines in serum were determined.The study found that SSW can effectively reduce the diarrhea index(P<0.05)and reduce the high sensitivity of intestinal tract(P<0.05)of IBS-D rats.SSW can significantly alleviate the inflammatory reaction,gastrointestinal motility and digestive dysfunction of the IBS-D rats and the increased sensitivity of the intestine.The principal component analysis(PCA),the principal co-ordinates analysis(PCoA)and the non-metric multidimensional scale analysis(NMDS)based on the Beta diversity distance showed that there were significant differences in the composition of the gut microbiota among the five groups(P < 0.05).The abundance,uniformity and diversity of the model group were the lowest.Compared with the control group,the model group showed a significant increase in Proteobacteria,Actinobacteria,Veillonococcus and Mycoplasma(P<0.05),and Pleaverella was significantly reduced(P<0.05).Compared with the model group,the Proteobacteria and Mycoplasma of SSW were significantly reduced(P<0.05),and Clostridium,Turicibacter and Romboutsia were significantly increased(P<0.05).Thisstudy shows that SSW plays a role in the treatment of IBS-D by partially regulating the structure of the intestinal flora,and there is a synergy between ESW and WWZS.
